Genotyping of the Caprine Kappa-casein Variants in Egyptian Breeds

Abstract: This study aimed to detect the genetic variants of kappa-casein (k-CN) in four breeds of goat reared in Egypt using PCR-RFLP technique. Three primers 13F, kb1 and kb2 with two restriction enzymes Alw44I and HaeIII have been used for genotyping k-CN exon 4. The PCR product -resulted from kb1and kb2 primers- with size 459 bp was digested by Alw44I endonuclease to detect A or B and C alleles. The 645 bp fragment resulted from PCR with kb2 and 13F primers was digested by HaeIII for detecting A and E alleles. Allele B was identified by combining the results of digestion with Alw44I and HaeIII endonucleases. The results showed that the allele B is the most common allele in four tested breeds and appeared at a frequency 68.2%, whereas allele C was appeared with 22.7%. Allele A was displayed in different frequencies ranged from 10 to 45.5%. The appearance of each allele was in homozygous or heterozygous genotypes. Allele E was not present in all tested animals. Identification of the genotypes of Egyptian goat milk protein will enable us to establish better strategy for breeding the Egyptian goat contributed in filling the nutritious gap in milk production.
