  To date, the search for novel pharmacotherapy from medicinal plants for psychiatric illnesses has significantly progressed. The present study was performed to evaluate the anxiolytic and antidepressant like activities of the K.parviflora rhizome extract. Aged male Wistar rats were orally administered the alcoholic extract of this plant at various doses ranging from 100, 200 and 300 mg kgˉ1 BW once daily for 7 days. The anxiolytic and antidepressant activities were performed after both single and repetitive treatment for 7 days using elevated plus maze and forced swimming tests respectively. The results showed that the extract decreased immobility time with the increase swimming time. However, no changes in number of open arm entries and time spent in open arm were observed. These results suggested the anti-depression activity of the plant extract. Therefore, K.parviflora may be served as a potential resource for natural psychotherapeutic agent against depression. However, further studies were still required.

  Problem statement: At present, the development of protective strategy against Alzheimer’s Disease (AD) is increasing its importance due to the high prevalence of AD, a limitation of therapeutic efficacy and its high impacts on economic and social aspects. The development of the preventive and therapeutic strategy to protect against the path physiology induced by free radicals in AD from antioxidant has gained very much concentration. Quercetin, one of the flavonoids in fruits and vegetables, has a powerful antioxidant activity both in vitro and in vivo. However, poor absorption, rapid metabolism and limited ability to cross the blood-brain-barrier are obstacles to its use for treatment of AD. Liposome’s have been used as an effective delivery system to the brain. Advantages associated with the nasal administration over oral route include higher bioavailability due to no first pass hepatic metabolism and rapid absorption leading to shorter time to onset of effect. Based on all these points, the possible effects of quercetin liposomes via nasal route on improving cognitive behavior and neurodegeneration in animal model of Alzheimer’s disease were investigated. Approach: Male Wistar rats were pretreated with quercetin liposome’s, containing 0.5 mg of quercetin in 20 μL (dose = 20 μg), via intranasal route once daily continually for 2 weeks before and 1 week after AF64A administration. Learning and memory was evaluated using the Morris water maze test at 7 days after the AF64A administration and then the rats were sacrificed for determining the density of neurons and cholinergic neurons in hippocampus using histological and immunohistochemical techniques. Results: Nasal administration of quercetin liposome’s significantly prevented changes of spatial memory of AF64A treated rats. The cognitive enhancement of quercetin liposome’s was found to be related to its ability to inhibit the degeneration of neurons and cholinergic neurons in hippocampus. Conclusion: Nasal administration of quercetin liposome’s possessed cognitive enhancer and neuroprotectant in animal model of AD, suggesting a potential novel therapeutic strategy against AD of quercetin liposomes.

  Problem statement: Stroke or cerebral ischemia has been recognized as one important problem worldwide. To date, the effectiveness of protective and therapeutic strategies against stroke is still very limited. Therefore, the development of novel strategy is required. Porcine brain is traditional believed to improve brain functions. Recent studies showed that the extract of porcine brain could protect against brain damage related to the oxidative stress, therefore, we hypothesized that it could protect against brain damage in stroke. Approach: To test the potential of porcine brain extract as the novel protective supplement against stroke, various doses of porcine brain extract at doses of 0.5 and 2.5 mg kg-1 b.w. were orally given to male Wistar rats, weighing 300-350 g, at the period of 14 days before and 21 days after the occlusion of right middle cerebral artery. Then, all rats were determined the neurological score, motor performance, cognitive function and brain infarct volume. Moreover, the possible neuroprotective mechanisms of the extract were also determined via the alteration of Malondialdehyde (MDA) or lipid peroxidation product and via the activities of scavenger enzymes including Superoxide Dismutase (SOD), Catalase (CAT) and Glutathione Peroxides (GPx). Results: The results showed that the low dose of porcine extract decreased the infarct volume and improved brain functions including neurological score, motor performance and memory deficit. In addition, it also decreased MDA but increased the activities of SOD, CAT and GPx. Conclusion: Our results suggested the potential role of porcine brain extract as neuroprotectant. The possible underlying mechanism appeared to be related to the enhanced activities of SOD, CAT and GPx which in turn resulted in the decrease MDA. Moreover, our findings may shed light on the pharmacologic basis for the clinical application of traditional Chinese medicine to protect against stroke.

  Problem statement: Demand for cognitive-enhancing drugs is growing. Numerous medicinal plants possessing antioxidant activity have received much attention as food supplement to improve cognitive function. Quercetin is a potent free radical scavenger and antioxidant. However, the limitations of quercetin: Rapidly metabolized is an obstacle to its use for a cognitive enhancer. In addition, the burden of blood brain barrier can be overcome by nasal administration and liposomes. In the present study, we investigated whether nasal administration of quercetin liposomes could improve spatial memory in healthy adult rats. Approach: Male Wistar rats were pretreated with quercetin liposomes, containing 0.5 mg of quercetin in 20 μL (dose = 20 μg), via right nasal cavity once daily continually for 4 weeks. Evaluation of rodent learning and memory was assessed by Morris water maze test and then all rats were sacrificed for determining the survival and cholinergic neurons densities in hippocampus. Results: Quercetin liposomes via nasal route treated rats exhibited a significant improvement in cognitive performance. In addition, nasal administration of quercetin liposomes also resulted in induced the densities of survival and cholinergic neurons in hippocampus. However, further researches about the precise underlying mechanism are still required. Conclusion: Our studies demonstrate that quercetin liposomes via nasal administration may have a candidate for cognitive enhancer in the future.

  Problem statement: Although the global prevalence of psychological disorders is increasing continually, the efficacies of therapeutic strategies are still very limited. Oxidative stress has been implicated in the pathogenesis of psychiatric disorders and substance possessing antioxidant activity can alleviate the mentioned conditions. Therefore, we hypothesized that Kaempferia parviflora, a medicinal plant claimed for nerve tonic, also possessed the neuropharmacological activities which provide the advantage for psychiatric disorders. Approach: Adult male Wistar rats, weighing 180-220 grams, were orally given the ethanolic extract of K. parviflora at doses of 150, 200 and 250 mg kg-1 BW once daily for 2 weeks. Then, they were determined the neuropharmacological activities including anxiolytic, anti-depression like behavior and cognitive enhancement after single dose, 1 and 2 weeks of treatment. Results: K. parviflora possessed anti-depression like behavior and cognitive enhancement at all treatment duration. Unfortunately, this substance failed to show anxiolytic like activity. Conclusion: Our results suggested that K. parviflora might be used as a novel therapeutic strategy for psychiatric disorder and cognitive enhancement. However, further investigations about precise underlying mechanism are still required.
