In this study, we established an efficient Agrobacterium-mediated transgenic system in rice (Oryza sativa L.). During the studies in the japonica (Xiushui-11) and indica rice cultivars (XC-95), the role of various factors responsible for virulence of Agrobacterium was screened more carefully to establish a better transformation system for the rice cultivars. MS and AB media (as VIM) proved to be efficient for Xiushui-11 and XC95, respectively. 100 μM AS (acetosyringone), pH 6.0 (VIM and COM) and use of either sucrose, glucose (Xiushui-11) or sucrose (XC-95) produced the best results for both of japonica and indica rice. Bacterial cell density A600 0.4-0.6 for 10-15 min was the most favorable optimized condition. Completely dark (Xiuishui-11) and light (XC-95) conditions during co-cultivation period were found to be ideal for transformation process. The significance of achieved results has been discussed properly. |

NP levels of 120:60, 180:80 and 240:100 kg ha-1, seed rates of 100, 125 and 150 kg ha-1 and row spacing of 15, 30 and 45 cm were studied in the experiment. Tillers m-2, plant height, thousand grain weight, grain yield and biological yield were significantly affected by NP levels while tillers m-2 and plant height of wheat were significantly affected by seed rates and row spacing as well. |

The effect of tillage operations, one plough (minimum tillage), two plough (conventional tillage) and four plough (maximum tillage) and seed rates, 75, 100, 125, 150 and 175 kg ha1 on wheat were studied. Statistical analysis of the data revealed that different tillage operations had significantly affected tillers/m2, thousand grain weight, number of grains/spike and grain yield. Maximum tillers/m2 (305), thousand grain weight (43.2 g), number of grains/spike (57) and grain yield (6266 kg ha1) were obtained from plots ploughed four times. Maximum tillers/m2 (324), grains per spike (59), thousand grain weight (42.9) and grain yield (6266 kg ha1) were recorded at seed rate of 100 kg ha1. Interaction between tillage practices and seed rate had a significant effect on tillers/m2, thousand grain weight, number of grains/spike and grain yield of wheat. As interaction between tillage operations and seed rates was significant therefore separate regressions were fitted to the data and xmax was calculated from the equation Y = a+bx+cx2 for each parameter and treatment as -b/2c. |
