  Tolaki chicken is a kind of Indonesian local chickens, that belong to the ability of anti viral responses. This ability is controlled by the present of antiviral Mx (myxovirus resistance) gene. The Mx gene codes for a protein with antiviral activity. The objective of the study was to prove the tolaki chicken Mx gene genotype is associated with antiviral and production traits. Mx/Hpy 81 gene was genotyped in 103 tolaki chickens with PCR-RFLP. A total of 30 chickens were challenged with ND gen VIIb virus (104 CLD50)/chicken. PCR was used to amplify genomic DNA for Mx gene (299 bp). The amplimer was cut by Hpy 81 produce three genotypes: AA, AG and GG and two alleles: A allele (299 bp) and G allele (200 bp and 99 bp). Frequency of A allele (0.74) was higher than G allele (0.26). The all parameters of production traits in challenge test group were not significantly different in AA, AG and GG genotypes. The daily weight gain, feed intake and FCR were significantly different in AA, AG and GG genotypes of chickens control group. The parameters of antiviral traits showed that vitality of AA (50%) and AG (50%) of chickens were better then GG (10%) in challenge group. The vitality of AA (100%) and AG (100%) were better GG (33.33%) in control group. The study postulated that Mx gene genotype could be associated with production and antiviral traits in tolaki chicken. AA and AG genotype are more resistant and show better production than GG genotype.

  Background: The Mx gene plays a crucial role in the antiviral responses of chicken. The Mx gene codes for Mx protein, which possesses antiviral traits. The non-synonymous G/A single nucleotide polymorphism (SNP) at position 2032 of chicken Mx cDNA results in a change at amino acid 631 of the Mx protein. This mutation affects the antiviral activity of the Mx molecule. Objective: The aim of the study was to evaluate the genetic polymorphism of the Mx gene in the native chickens of South East Sulawesi, Indonesia, using PCR-RFLP. Materials and Methods: The Mx gene was genotyped in 25 Tolaki chickens from each of Konawe Regency and South Konawe Regency and in 21 Kampong chickens from Kendari City. Tolaki chicken is traditionally used as a medium for medicinal treatments in the Tolaki culture. PCR was used to amplify genomic DNA for the Mx gene (299 bp). The amplifier was cut using the Hpy81 enzyme. Results: The genotyping of the Mx gene of native chicken produced three genotypes, AA, AG and GG and two alleles, A (299 bp) and G (200 bp and 99 bp); the frequency of A was higher than that of G. The value of x2 showed that Mx|Hpy81 was in Hardy-Weinberg equilibrium. The values of He and PIC were 0.47-0.49 and 0.36-0.37, respectively. Conclusion: These results indicated that Mx|Hpy81 gene was polymorphic in all strains of chicken that were genotyped. The Mx|Hpy81 gene demonstrated a high potential for use as a genetic marker for resistance to Avian influenza and Newcastle disease infection in Indonesian native chickens.

  Background and Objective: Kampung chickens are native Indonesian chickens that have the ability to utilize the low quality of local feed. This research was aimed to study the responses of kampung chickens to local-based feedstuff. Materials and Methods: A total of 140 Tolaki strain kampung chickens (25 females and 5 males) were raised to obtain production performance. Chickens were divided into two equal groups (a group of 70 receiving local feed and a group of 70 receiving commercial feed). The local feedstuffs used in this research were yellow corn, rice bran, fish meal, banana peel meal and papaya leaf meal. The obtained data was analyzed using a t-test (Independent samples t-test). The research variables were indicators of production performance, including feed intake (FI), average daily weight gain (ADG) and feed conversion ratio (FCR). Results: Research results showed that there were highly significant correlation (p<0.01) between the body weight gain and feed consumption. However, the different feed treatments did not contribute differences (p>0.05) in FCR. Conclusion: The local feedstuff for kampung chickens could provide a benefit as a substitute for commercial feed because the local feed had a good impact on chicken production performance.
