  The development of instruments to assess substance use that are easy to use, valid and reliable across cultures is an important task. The present study investigates the reliability and validity of the Chinese language version of the SDS (SDS[Ch]) when used to measure severity of dependence among heroin users in Taiwan (n=522). Data were collected on demographic data, heroin use behaviours, and criminal convictions. Taiwanese heroin users recorded high SDS[Ch] scores and the results support the validity and reliability of the Chinese version of the SDS. A positive correlation was found between SDS[Ch] scores and DSM-IV criteria for heroin dependence. SDS[Ch] scores were positively related to heroin injection, frequency of heroin injection, spending on heroin, earlier age of onset of heroin use and more drug-related criminal convictions. The findings support the suggestion that the concept of dependence as assessed by the SDS has cross-cultural validity.

  It is known that the SREBP1c gene is an important gene responsible for adipogenesis and regulation of the expression of genes controlling fatty acid biosynthesis. Its expression levels increase in parallel with obesity. Therefore, the present study focused on screening the genetic variation within bovine SREBP1c gene and analyzing its effect on growth traits in 1035 individuals belonging to four Chinese cattle breeds (QC, NY, JX, CH) using PCR-SSCP, DNA sequencing, and forced PCR-RFLP methods. The results revealed two novel mutations: NC_007317: g. 10781 C > A (457aa), 10914 G > A (502aa). Association analysis with growth traits in the Nangyang breed indicated that: The SNPs in the bovine SREBP1c gene had significant effects on body weight and average daily gain at birth, 6 and 12 months old (P < 0.05 or P < 0.01). Therefore, these results suggest that the SREBP1c gene is a strong candidate gene that affects growth traits in cattle.

  Myocyte enhancer factor 2A (MEF2A) gene encodes a member of the myocyte enhancer factor 2 (MEF2) protein family, which involves in vertebrate skeletal muscle development and differentiation. The important function of MEF2A implies this gene is a potential candidate gene for considering association analysis between polymorphism and growth trait. Sequencing and forced-PCR-RFLP were used to analyze the polymorphisms of the MEF2A gene in 1009 cattle from three Chinese indigenous breeds. Three SNPs (C1598T, G1641A and C1734T) were detected in the cattle MEF2A gene, leading to a missense (P420L) and two silent mutations (E434 E, P465P). The frequencies of 1598CC genotype and the frequencies of allele 1598C varied from 0.884-1.000 in the analyzed population (NY, QC and JX). The genotypic and allelic frequencies of G1641A and C1734T in three populations were 0.034-1.000. The C1598T was associated with body length at 6 and 12 months of cattle. The G1641A was associated with average daily gain at 12 months of cattle. And the C1734T affected the average daily gain and body weight at 6 months of cattle. Those three mutations affected the early growth and development of cattle, which could be potential molecular makers in assistant selection of cattle breeding.

  The goal of this study was to examine the effect of maize plumule extracts on mammary gland tissue and serum hormone levels in SD rats suffering from mammary gland hyperplasia and to investigate the mechanism by which maize plumule extracts inhibit mammary gland hyperplasia. About 56 rats were subcutaneously injected with progesterone combined with estradiol benzoate for the induction of mammary gland hyperplasia and were then randomly divided into 7 groups. Maize plumule extracts were subsequently given to the rats at doses of 0, 1.25, 2.5, 5, 10, 20 and 40 mg kg-1 daily for 20 days. Following treatment, hormone levels in the blood were measured and the morphology of the rat mammary glands was observed by microscopy. Results showed that high doses of maize plumule extracts significantly improved the morphology of lesional tissue and significantly reduced the levels of Follicle-Stimulating Hormone (FSH), serum Estradiol (E2) and Testosterone (T) in the serum and increased the levels of Prolactin (PRL) and serum Luteinizing Hormone (LH). However, the treatment did not have a substantial effect on serum Progesterone (P) levels. It can be concluded that maize plumule extracts containing rich adenine derivatives administered at doses of 2.5-40 mg/kg/day could effectively prevent and treat rat mammary gland hyperplasia.

  The results of analysis of context-dependent codon bias designed to identify the relationship between codon bias model for the context surrounding the cleavage site and the model for the corresponding cleavage site, applied to genome data form 66 isolates of Foot and Mouth Disease Virus (FMDV) representing all seven serotypes were reported. The cleavage sites in the polyprotein were identified from all parts of the FMDV genome and the similarity of Codon Bias Model for regions flanking the cleavage sites was estimated. Although, the two amino acid residues which was comprised of the cleavage site were not conserved, one residue always tended to be conserved, suggesting that the conserved residue was essential for the cleavage process. In addition, although the similarity of Codon Bias Model for some positions in the most target contexts was ensured, the scale of similarity of Codon Bias Model was larger in the regions flanking the 2A/2B, 2C/3A, 3A/3B, 3B/3C and 3C/3D sites than that of the rest. Furthermore, correlation analysis between the Codon Bias Model for each position in the target context surrounding the cleavage site and that of the corresponding site was performed it was concluded that the significant relationship existed in the VP4/VP2, 2A/2B, 2B/2C and 3A/3B sites, respectively. The interesting phenomena probably suggested that context-dependent codon bias surrounding some cleavage sites was existence in FMDV and the Codon Bias Model for these regions played potential roles in shaping specific characteristics of these corresponding sites at the level of transcription or translation.

  Six 5-coordinate 2,6-bis(imino)pyridine metal complexes, [2,6-(ArN=CMe)2C5H3NMCl2 · nCH3CN] (Ar = 4-MeC6H4, M = Zn, n = 0.5, Zn1, M = Cd, n = 1, Cd1; Ar = 2,6-Et2C6H3, M = Zn, n = 0.5, Zn2, M = Cd, n = 0.5, Cd2; Ar = 2,4,6-Me3C6H2, M = Zn, n = 1, Zn3, M = Cd, n = 1, Cd3), were synthesized in acetonitrile by the reactions of the corresponding bis(imino)pyridines with ZnCl2 or CdCl2 · 2.5H2O, respectively. The structures of Zn1-Zn3 and Cd1-Cd3 were determined by the single-crystal X-ray diffraction. In all complexes, the ligand is tridentate with further coordination by two chlorides, resulting in a distorted trigonal bipyramid. All complexes self-assemble through hydrogen bonding interactions to form a 3-D supramolecular structure. At 298 K in dichloromethane, all complexes have blue luminescent emissions at 405-465 nm, which can be attributed to ligand-centered π* → π transitions. The zinc and cadmium centers play a key role in enhancing fluorescent emission of the ligands.

  We have previously shown that ASK1-interacting protein 1 (AIP1) transduces tumor necrosis factor-induced ASK1-JNK signaling. Because endoplasmic reticulum (ER) stress activates ASK1-JNK signaling cascade, we investigated the role of AIP1 in ER stress-induced signaling. We created AIP1-deficient mice (AIP1-KO) from which mouse embryonic fibroblasts and vascular endothelial cells were isolated. AIP1-KO cells show dramatic reductions in ER stress-induced, but not oxidative stress-induced, ASK1-JNK activation and cell apoptosis. The ER stress-induced IRE1-JNK/XBP-1 axis, but not the PERK-CHOP1 axis, is blunted in AIP1-KO cells. ER stress induced formation of an AIP1-IRE1 complex, and the PH domain of AIP1 is critical for the IRE1 interaction. Furthermore, reconstitution of AIP1-KO cells with AIP1 wild type, not an AIP1 mutant with a deletion of the PH domain (AIP1-ΔPH), restores ER stress-induced IRE1-JNK/XBP-1 signaling. AIP1-IRE1 association facilitates IRE1 dimerization, a critical step for activation of IRE1 signaling. More importantly, AIP1-KO mice show impaired ER stress-induced IRE1-dependent signaling in vivo. We conclude that AIP1 is essential for transducing the IRE1-mediated ER stress response.

  Sixteen crossbred barrows (Duroc X Landrace X Yorkshire) weaned at 28 days with an initial body weight of 5.12-8.64 kg and no antibodies to PCV2 were used in a 5-week trial to evaluate the immune response after intranasal inoculation with type-2 porcine circovirus. The percentage of peripheral blood ANAE+ T cells, PCV2 antibody titer, immunoglobulins, and serum IFN-γ, IL-2, and sIL-2R levels were determined. The results show that the percentage of peripheral blood ANAE+ T cells was notably higher in inoculated pigs 5 days post-inoculation (dpi) and significantly lower 21 dpi than in the control group. The anti-PCV2 titer in the inoculated group was significantly higher than that in the control group 14 dpi. The immunoglobulin profile of the 2 groups also differed. The IFN-γ level in the inoculated group decreased significantly from 7 to 28 dpi, as compared with the control group. The results suggest that PCV2 challenge results in an immune system response.
