  The control region (D-loop) of Anabarilius grahami was amplified by PCR amplification with a pair of specific primers. The sequence with the complete nucleotide control region from A. grahami mitochondrial was cloned and directly sequenced. The length of this region (D-loop) contained 930 bp nucleotides and the T, C, A and G contents were 29.8%, 22.5%, 33.0% and 14.7% respectively. The mtDNA control region of A. grahami could be partitioned into three domains, namely, the termination associated sequence domain, the central conserved sequence domain and the conserved sequence block domain. The extended termination associated sequence (ETAS), two conserved blocks (CSB-F, CSB-D) in the central conserved sequence block domain and three conserved sequence blocks (CSB1, CSB2, CSB3) in the conserved sequence block domain were successfully identified and their homology compared with other fish. The genetic diversity analysis of A. grahami from the Endangered Fish Breeding Center (EFBC) of the Kunming Institute of Zoology (KIZ), Mingxing Fish Cave (Jiangchuan), Niumo Village (Jiangchuan) was analyzed. The result indicated a very low genetic divergence between two natural populations, and the genetic diversity of the population from EFBC was much higher; they had recovered better than others.

  A new species of the genus Protocobitis, was discovered in an underground water source (110 m depth) located about 5 km away from the county town of Wuming, Guangxi, China, in May 2006. The new species, Protocobitis polylepis sp. nov., is distinguished from the single congener by the following characteristics: 1) light black pigmentation present vs. no pigmentation; 2) entire body covered with scales except for its head and abdomen vs. rudimentary scales present only along the midline of the body; 3) head length 24.1%-24.8% of SL vs. 19.8-22.1% of SL; 4) body depth 16.2%-16.3% of SL vs. 11.5%-13.0% of SL; 5) Inner rostral barbel length 19.8%-21.0% of head length vs. 9.4%-11.8% of head length; outer rostral barbel length 28.6%-30.2 % of head length vs. 15.3%-21.8% of head length; maxillary barbell length 44.6%-46.0% of head length vs. 22.4%-31.8% of head length.

Field surveys were conducted in the freshwater ichthyfauna of the Liu River and the Hongshui River in the Hechi Prefecture, Guangxi Province between 1994 to 2008. According to the field surveys and reference records, the two rivers harbor a total of 185 species belonging to 9 orders, 21 families, and 107 genera. Compared with previous investigations, 21 species were newly discovered in this area, including 1 new genus and 19 new species. There is a total of 134 fish species in the Liu River, under 8 orders, 19 families, and 88 genera. The species of Cypinifomes has the highest number in popoulation. The Perciformes came in second, and the Siluriformes is the third. The numbers of species for other orders are comparatively smaller than the three orders, only 1-3 species. The total number of species in the Hongshui River is 157, belonging to 8 orders, 20 families, and 101 genera. The number of species for Cypinifomes is the most. The Siluriformes takes second place, and the Perciformes is the third. The numbers of species of other orders are comparatively smaller than the three orders. The fish diversity of Hongshui River is richer than Liu River and the reasons are that Hongshui River flows through such regions that karst and underground rivers are developed and drainage areas are broad, with plenty of water.

A nemacheiline loach Yunnanilus jinxiensis sp. nov. has been discovered in collections from a Pearl River Drainage in Ludong village, Jingxi county, and Guangxi of China in May 2008. Yunnanilus jinxiensis is a new member of the Yunnanilus pleurotaenis group. It can be distinguished from all other congeners in this group by the following characters: body entirely covered by scales, except between pectoral fins and pelvic fins; lateral line present, with 15–20 pores; dorsal fin with 8 branched rays; pectoral fin with 13–14 branched rays; the air bladder has two chambers, the anterior ones into bony capsules and a large free posterior chamber, filling body cavity; caudal fin slightly emarginated; body depth 3.9–4.6 times the standard length (SL); head length 4.1–4.4 times SL; caudal peduncle depth 7.5–9.0 times SL; eye diameter 5.5–6.7 times the head length (HL); interorbital width 2.2–2.4 times HL.

  In Nov 2006 and Oct 2008, members of the Kunming Institute of Zoology (KIZ) took surveys of fishes in the Niulan River, a tributary of Jinsha River, Yunnan, and collected 42 fish species. According to the historical record and the fish specimens preserved in the collection room of KIZ, there are a total of 59 fish species in Niulan River, which belong to 46 genera, 12 families and 5 orders. Ichthyologic fauna of Niulan River were separately analyzed by traditional sorting method and the Value of Faunal Presence (VFP) method at the respective levels of order, family and genus. Two kinds of different ranking methods get two rather different results. Traditional sorting analysis indicated that some world-distributed families and genera had a small proportion in the Niulan River. On the contrary, the VFP of some endemic families and genera turned out to be relatively high. The families Amblycipitidae and Channidae have two genera and which distribute in Africa and south Asia, with the highest VFP. The genera Jinshaia and Metahomaloptera, which distribute in the Yangtze River, have a VFP of 100%. Pseudogyrinocheilus and Oreias have one species, with a VFP of 100%. The VFP method can reflect better the character of ichthyologic fauna than the traditional sorting method. By combination of these two methods, we analyzed the ichthyologic fauna of Niulan River, and then we compared it with the lower reaches of Jinsha River ichthyologic fauna from Shigu to Yibin, which indicated high comparability.

  Based on the online bioinformatics analysis and alignment results, three RNAi sequences target to the Mus musculus Bicc1 gene were obtained. The three interference fragments were synthesized and cloned into pRS-Hush shRNA Vector. The Bicc1 eukaryotic expression vector pEGFP-C3-Bicc1 was constructed, tagging the GFP to the N-terminal of the Bicc1 protein. The pEGFP-C3-Bicc1 and three pRS-Hush-RNAi were co-transfected into the cultured HEK-293 cells line, respectively. The two RNAi (pRS-Hush-RNAi-Bicc1-N/-C) that could knock-down the Bicc1 expression levels in HEK-293cells significantly were confirmed by cell immunofluorescent staining, semi-quantitative PCR and Western blotting. The results demonstrate that we have successfully obtained two efficent Bicc1 RNAi sequences, which lays a foundation for further studying on the construction of Bicc1 knock-down stable cell lines and biological function of mouse Bicc1 product.
