  Dibenzothiophene (DBT) and its derivatives can be microbially desulfurized by enzymes DszC, DszA, and DszB, which are encoded by the operon dszABC and contribute to the conversion in tandem. We investigated the expression characteristics of the dsz operon. Our results revealed that the levels of transcription and translation of dszA, dszB, and dszC decreased according to the positions of the genes in the dsz operon. Furthermore, the translation of dszB was repressed by an overlapping structure in the dsz operon. In order to get better and steady expression of the Dsz enzymes and optimize the metabolic flux of DBT, we rearranged the dsz operon according to the catalytic capabilities of the Dsz enzymes and expressed the rearranged dsz operon, dszBCA, in Rhodococcus erythropolis. After rearrangement, the ratio of dszA, dszB, and dszC mRNAs in the cells was changed, from 11:3.3:1 to 1:16:5. Western blot analysis revealed that the levels of expression of dszB and dszC had been enhanced but that the expression of dszA had decreased. The desulfurization activity of resting cells prepared from R. erythropolis DRB, which carried the rearranged dsz operon, was about 12-fold higher than that of resting cells of R. erythropolis DRA, which carried the original operon in a similarly constructed vector.

  The present study was designed to determine the effects of resveratrol supplementation on exercise-induced apoptosis of rat hepatocytes. The rats were divided randomly into the sedentary group (Group I) and exhaustive exercise groups (Group I-IV). Each group contains 12 animals. The rats of exhaustive exercise groups received daily oral administration of resveratrol at doses of 0, 200 or 400 mg/kg/day for 30 days. The rats of sedentary group received daily oral administration of vehicle only (10 mL/kg/day) for the same period. Exhaustive exercise was performed on a motor-driven rodent treadmill. Paraffin-embedded liver sections were stained using the Streptavidin Peroxidase (SP) immunohistochemistry technique for BAX and Bcl-2 detection. The results showed that exhausting exercises could make the expression of BAX proteins in the liver and BAX/Bcl-2 ratio dramatically increases, the expression of Bcl-2 proteins obviously decreases. After resveratrol supplementation, the expression of BAX proteins and BAX/Bcl-2 ratio obviously decreased, the expression of Bcl-2 proteins obviously increased. Which indicated that resveratrol supplementation can inhibit exercise-induced apoptosis of rat hepatocytes.

  Tao-Hong-Si-Wu-Tang (THSWT) is a famous traditional Chinese herbal medicine formula which has traditionally been used in China for about 1000 years. The previous studies have shown that THSWT had anti-fatigue effects but the mechanisms are scanty. The present study was designed to evaluate anti-fatigue mechanisms of THSWT through determining the change of glucose and lipid metabolism. The rats were randomly divided into four groups: Sedentary Control group (SC); Exercise Control group (EC): Sedentary plus THSWT Treated group (STT) and Exercise plus THSWT Treated group (ETT). Rats in the STT and ETT groups received 20 mL kg-1 BW of THSWT solutions. After 28 days, rats in the EC and ETT groups were forced to swim for 90 min with a load (5% BW). The results showed that THSWT could improve blood glucose levels of rats during exercise as well as increase the liver and muscle glycogen contents while decreasing the plasma Free-Fatty Acid (FFA) levels. The anti-fatigue mechanisms of THSWT might be related to the improvement in the metabolic control of exercise and the activation of energy metabolism.
