  Wheat is most widely cultivated cereal and important source of food and protein worldwide. Wheat is a staple diet containing most of the ingredient, for wide range of food. The unique property of wheat flour is to form dough and this property is due to gluten required for making preparation of chapatti and bread. Fractionation and characterization of seed storage proteins of fourteen wheat varieties cultivated in sindh was carried out by Sodium Dodecyle Sulphate Polyacrylamide Gel Electrophoresis (SDS-PAGE). Wheat grains of fourteen (14) varieties were collected from different region of sindh (Pakistan). Electrogram of each variety were calculated by Jaccard’s Similarity Index (JSI). Genetic diversity of different sindh wheat varieties was measured by constructing dendogram of High Molecular Weight (HMW) and Low Molecular Weight (LMW) gluten sub unit bands. The dendrogram profile data showed the variation in the number and position of bands from one variety to the other, while some bands are common.

  SDS-PAGE electrophoresis is the most effective as well as one of the simplest methods for separating proteins. The extraction and fractionation technique was applied to characterize rice seed proteins. Identification and screening of rice proteins carried out from 14 varieties of rice collected from Nuclear Institute of Agriculture (NIA) Tandojam. Rice seed proteins were extracted with phosphate buffer for total proteins. After quantification, proteins were separated by SDS-PAGE Electrophoresis. Analysis of rice seed proteins was performed on the results of SDS-PAGE using gel documentation system (Biorad) to find out the molecular weight of different varieties of rice followed by cluster analysis using SPSS to find out the diversity among different rice varieties.

  Protein is not only essential compound of animal but it is also important constitute of plants. Plants are good source of proteins. These proteins are mostly analyzed and identified from the fruits, seeds or any other parts of plants which are mostly used in diet but there are many other types of protein which are synthesized by other parts also, if one can know about synthesis of those proteins at different growth periods of different parts of plant then one may get interesting results. In this study stem of two plants were used for the analysis of protein i.e Pea (Pisum sativum) and Gram (Cicer arietinum). Molecular weight of protein was analyzed by SDS/Polyacrylamide Gel Electrophoresis and protein was stained with Lawsone dye by paper chromatography. Lawsone dye is protein dye which is mostly used for the staining of hair, skin and nails protein but in this research same dye was used for the staining of plant protein through paper chromatography.
