  Insulin-like Growth Factor-I (IGF-I) is a very important candidate gene which generates several IGF-I mRNA variants. Because of its alternative splicing this gene is involved in very complex biological functions such as the mediation effect of promoting the growth activities of growth hormone. The mammalian IGF-I gene contains 6 exons of which exon 1 and 2 produced various leader sequences and exon 5 and 6 produced different E domain by different splicing forms. In this study, the 3′ends sequences of IGF-I messenger ribonucleic acid (mRNA) in Chinese red steppes liver were cloned by 3′-Rapid Amplification of cDNA Ends (3′RACE) technique. Researchers found that there are two types of 3′ends sequences containing exon 5 (Eb) and 6 (Ea) in Chinese red steppes. According to the result of 3'RACE and the reports about leader exons, we have assembled the possible transcripts; type Ea of class 1, type Eb of class 1, type Ea of class 2 and type Eb of class 2. Semi-quantitative RT-PCR was used to identify the distribution of these four new transcripts of IGF-I mRNA expression in Chinese red steppes. The results show that type Ea of class 1 was expressed in all tissues tested whereas, the type Eb of class 1, the type Ea of class 2 and the type Eb of class 2 were expressed in minority tissues examined. The study shows that class 1 IGF-I mRNA has higher expression than class 2 and that type Ea of class 1 is expressed in the highest mRNA level.

  MicroRNAs (miRNAs) are endogenous small regulatory RNA molecules that play a crucial role in the regulation of gene expression in eukaryotes. Recent studies indicate that miRNAs are mechanistically involved in mammalian spermatogenesis however, little is known regarding their expression patterns during porcine testis development. Here, researchers characterized the spatio-temporal expression patterns of two miRNAs, miRNA-122 (miR-122) and miRNA-221 (miR-221) in porcine diverse tissues and four stages of testis development by using a Stem-Loop Quantitative RT-PCR Method. The result showed that miR-122 was relative highly expressed in adult porcine testis among all tissues examined except for the liver and had an increasing expression pattern during porcine testis development from birth to adult. Conversely, miR-221 was relative lowly expressed in adult porcine testis among all tissues examined and had a decreasing expression pattern during porcine testis development. The results suggested that miR-122 and miR-221 may play distinct roles during testis development or spermatogenesis. In summary, determination of the spatio-temporal expression patterns of miR-122 and miR-221 in porcine developing testes provides a useful resource for further illustrates their likely roles in pig testis development or spermatogenesis as well as in related mammals.

  Insulin-Like Growth Factor 1 (IGF-1) is a single copy gene. It generates several IGF-1 mRNA variants due to alternative splicing. Spliced variants of IGF-1 being some small peptides derived from spliced variants of IGF-1 have critical roles in cell metabolism and growth of various vertebrate species. Porcine IGF-1 gene has two promoters and the existence of Exon 6 has been confirmed in Songliao Black pig in the laboratory. The aims of the present study were to identify new spliced variants of IGF-1 gene including Exon 5 and to investigate their expression profile in different tissues of Songliao Black pig. The cDNAs of two new spliced variants of IGF-1 gene were cloned by RACE. These two variants were named Class I IGF-1-Eb and Class II IGF-1-Eb, respectively. The Class I IGF-1-Eb contains an Open Reading Frame (ORF) of 564 bp encoding 187 amino acids (GenBank Accession No. FJ914497). The Class II IGF-1-Eb contains an ORF of 516-bp encoding 171 amino acids (GenBank Accession No. FJ914498). Tissue distributions of mRNAs of Class I IGF-1-Eb and Class II IGF-1-Eb were investigated in Songliao Black pig by semi-quantitative RT-PCR. Both of the new mRNA variants were expressed in a variety of tissues. Class I IGF-1-Eb mRNA was more abundant than Class II IGF-1-Eb mRNA in most tissues except for duodenum and longissimus muscle (p<0.05).
