  The gloshedobin, a snake venom thrombin-like enzyme was biosynthesized in the soluble form and purified by egg yolk antibody-coupled adsorbents from E. coli. As His-tag is not favored from the point of view of high-level and soluble expression, we herein constructed a recombinant gloshedobin without His-tag and developed a novel egg yolk antibody (IgY)-immunoaffinity chromatography for its purification in a higher activity yield. The purification process involving Octyl Sepharose FF, IgY-immunoaffinity chromatography and Source Q, yielded 454.7U mg-1 protein of interest and 34.8% activity recovery. The anti-gloshedobin IgY was obtained from eggs by injecting diluted gloshedobin into the breast muscle of laying hens and then purified by several steps including 3.5%, 8.5% and 12% polyethylene glycol-6000 precipitation and affinity chromatography using gloshedobin-coupled agarose gel. The obtained IgY was covalently linked to CNBr pre-activated agarose gel, Sepharose 4B FF, to yield immunoaffinity adsorbents. Both immunological and enzymatic activities of the purified enzyme were determined by western-blotting analysis and fibrinogen clotting assay, respectively.

  Internet sellers hope to let the consumers find their commodities easily, but there is a divergence between commodities presentation requirements and the limitation of a web page. In order to pursuit the profit-maximizing, a web page must contain the most welcome commodities. In this study, we proposed a multiple candidate set method to discover those most welcome commodities from users' browsing data. Our approach utilizes frequent dependent relationships among the commodities to partition the unique candidate set, composed of all single frequent item, into multiple sub-candidate sets for computing the frequent browsing patterns, which avoids the problem of searching through a large space of candidate set. Our experiments illustrate the benefits of the proposed method against the single frequent item candidate set.

  Identifying cliques with the same interests is valuable for online shopping which can make the recommendation and advertisements to target different users more accurately and maximize the benefits of advertisers, publishers and users. This study, has proposed an effective and efficient method to discover cliques for online shopping which firstly identifies clique leaders and clusters the most similar users, then computes clique cores among existing clique members and finally generates the complete cliques. A marked improvement is that two key factors, users’ behavioral characteristics and regular purchase information, are unified to discover cliques. This method can also remove effectively most of fake purchases through computing the operation similarity among different goods categories.

  Large amount of content with similar structures are being integrated into one big XML document, for example, the product records crawled from Web, on which users requires a quick retrieval. DOM tree is a memory model for HTML and XML documents. There are three basic queries on this model, (1) search content located by a given absolute path, (2) search content located by a given relative path, (3) get the path locating the given content. For an XML document with great width and depth, those queries often have a long response time because of time loss on tree traversal. It will be very helpful for Web applications and XML query processing to improve the query efficiency on DOM trees. Present study proposed three index structures to deal with the three basic queries, every of them can give a quick response for corresponding queries with backward search strategy. The experiments on different XML documents show that these index structures can reduce the query time effectively.

  A transactional system often has a steady distribution based on their decisions or responses on various transactions. If the system is changed, the distribution also often changes. It is a valuable work to detect anomaly caused by system changes based on different distributions. In this study, we modeled those decision or response signals into a series of time-related distributions and then proposed a method combining distance metrics and anomaly detection to discover whether changes have happened in some systems. Distance metrics on different distributions can decide whether changes have happened and anomaly detection can find what happened further. Extensive experiments show that our method has a good performance and can locate the anomaly accurately.

  Two resorcyl di[4-(4-alkoxy-2,3-difluorophenyl)ethynyl] benzoate liquid crystals (A10 and A12) were prepared and their mesomorphic properties were observed. A large polarisation value was obtained with longer terminal chains.

  Ultrafast optical Kerr effect of metallophthalocyanine-doped inorganic–organic materials was investigated using a femtosecond optical Kerr shutter at wavelength of 800nm. Experimental results showed that the dependence of the Kerr signals on the polarization angle between the pump beam and the probe beam could be controlled by changing the pump-probe intensity ratio. The pump-intensity dependence of the polarization characters of the Kerr signals probably arose from the contribution of light induced transient grating (LITG) to the Kerr signals.

  Here we identify a novel protein, named Parcs for pro-apoptotic protein required for cell survival, that is involved in both cell cycle progression and apoptosis. Parcs interacted with Apaf-1 by binding to the oligomerization domain of Apaf-1. Apaf-1-mediated activation of caspase-9 and caspase-3 was markedly decreased in a cytosolic fraction isolated from HeLa cells with reduced parcs expression. Interestingly, parcs deficiency blocked cell proliferation in non-tumorigenic cells but not in multiple tumor cell lines. In MCF-10A cells, parcs deficiency led to early G1 arrest. Conditional inactivation of parcs in genetically modified primary mouse embryonic fibroblasts using the Cre-LoxP system also resulted in the inhibition of cell proliferation. We conclude that Parcs may define a molecular checkpoint in the control of cell proliferation for normal cells that is lost in tumor cells.
