  Background and Objective: The roots of Codonopsis lanceolata have been used in traditional medicine for treatment of many diseases in China. Till now, there is no evidence on the liver protection effect of C. lanceolata on alcohol-induced liver injury. The purpose of this study was to investigate the hepatoprotective effect of steamed C. lanceolata (SCL) on ethanol induced liver injury in mice. Materials and Methods: Experimental mice were pretreated with different doses of SCL (100-400 mg kg1) for 2 weeks by gavage feeding. Biochemical markers and enzymatic antioxidants from serum, liver tissue were determined. Results: The results showed that the activities of ALT, AST and TG in serum, MDA level in liver tissue, decreased significantly (p<0.05) in the SCL-treated group compared with the alcohol group. On the contrary, the GSH level was increased markedly (p<0.05). Histopathological examination revealed that SCL (400 mg kg1) pre-treatment noticeably prevented alcohol-induced hepatocyte apoptosis and fatty degeneration. Moreover, LC-MS/MS analysis of SCL showed that it mainly contain Lobetyolin, Tangshenoside, Lancemasides F, Lancemasides B (Lancemasides E, Codonolaside), Lancemasides G, Lancemasides A (Codonoposide, Codonolaside) and Codonoposide. Conclusion: These results provide the evidence on possible application of SCL on acute alcohol-induced oxidative stress and liver damage in clinic.

  The objective of this study was to evaluate the effect of three cryorotectants (at three concentration each) and two cryopreservation protocols on the preservation of chicken Primordial Germ Cells (PGCs) from gonads at stage 19 and stage 28. The PGCs were cryopreserved using Dimethylsulphoxide (DMSO; 10%, 20%, 30%), glycol (GLY; 10%, 20%, 30%), polyethylene glycol (PEG; 10%, 20%, 30%). In a first series of experiments, we compared viability after three cryorotectants based protocol I and found the viability of PGCs showed very significant percentage (86.53%) (p<0.01) in a freezing media IV. We then compared an cryopreserve protocol I with 5% dimethyl sulfoxide (DMSO)+5% Glycol versus a cryoprotectant protocol II and observed a better viability with the former protocol (85.9% versus 67.4%, p<0.05). Finally, we compared viability of PGCs at three concentration each cryorotectant and found no significant difference p>0.05 between the concentration of 10% and 15% except freezing media III. When the concentration was 20%, the viability of PGCs was the lowest and showed significant difference p<0.05 or very significant difference (p<0.01) compared to other concentration. In conclusion, 5% DMSO+5%GLY with cryopreserve protocol I was the most effective cryopreservation for chicken primordial germ cells.

  This experiment was conducted to study the Polymorphism on exon 2 of THRSPα gene in goose. The Single Nucleotide Polymorphism (SNP) of (Thyroid Hormone Responsive Spot 14 gene) THRSPα gene was investigated in Zi geese, Wanxi White geese, Zhedong White geese and Shitou geese. One primer for exon 2 of THRSPα gene were designed according to the homologous sequence of duck. Six SNPs were found in the sequence of THRSPα gene and 6 genotypes AA, BB, CC, AB, AC and BC were detected in 4 goose populations. All the populations were in Hardy-Weinberg equilibrium at this polymorphic site (p>0.05). The results confirmed that there were polymorphisms in the exon 2 of THRSPα gene.

  The aim of the present study was to detect Single Nucleotide Polymorphism (SNP) in some areas of Pit-1 gene of goose to find genetic marker influencing on growth and carcass trait. The Single Nucleotide Polymorphism (SNP) of Pit-1 gene promoter was investigated in Wanxi geese. The primers for promoter in Pit-1 gene were designed according to the Pit-1 gene promoter of goose and the SNPs were detected by PCR-SSCP method. The results showed that only C-930T was detected for SNP and two genotypes (CC and TT) were found, respectively. The analysis of least square on genotypes and groups showed that there were significant differences (p<0.01) on body weight of different weeks in different groups and genotypes, however, there were no differences (p>0.05) on body weight of different weeks in genotype-group interaction effect, then, comparison of bodyweight in different groups show that the average body weight of wanxi white goose was the largest, the average body weight of taihu goose was larger than zi goose. Variance analysis on genotype suggested that the average body weight of CC genotype and TT genotype had significant differences (p<0.05) in 6, 8 weeks and had extremely significant differences (p<0.05) in 10 weeks. This study indicated that the polymorphic site could be a genetic marker for growth trait.

  In this study, the exons polymorphism of Growth Hormone (GH) gene were detected by Polymerase Chain Reaction-Single Strand Conformation Polymorphism (PCR-SSCP) and DNA sequencing method in 109 individuals from Shitou goose, Wanxi white goose and Zi goose. About 5 pairs of primer were designed according to the GH gene sequence of goose then the association of polymorphism of GH gene with the growth traits was studied. The results indicated there were two nucleotide mutations at P2 and P4 locus, respectively which resulted in 10 genotypes and 3 genotypes. The χ2-test showed that the genotype distributions of GH gene were in agreement with Hardy-Weinberg equilibrium in P2 and P4 locus (p>0.05), the difference was extremely significant in P2 locus on the distribution of genotype for Shitou goose and Zigoose while for Shitou goose, Wanxi white goose and Zi goose, it was extreme significant at P4 locus but there was no difference for the other locus. The least square on genotypes and groups analysis showed that there were significant differences (p<0.01) on growth traits in different groups however, only P4 locus have genotypes effect, the average value of AA genotype was the highest therefore, AA genotype could be the favorable marker for early breeding selection for high product trait of local goose.
