  In this experiment the screening material has been divided into three categories: I) Nine monosomic lines (1A, 1D, 2A, 2D, 3B, 4A, 4B, 5B, 7D) have shown resistance against powdery mildew, but still some plants were attacked by this disease. ii) Five monosomic lines (2B, 3D, 4D, 5D, 6A) have indicated susceptibility against this disease, but still some resistant plants were there. iii) The third group has included the monosomic line 7B, where all the plants were attacked by disease. The line 7B and the variety “poros” exhibited a 100 % susceptibility against powdery mildew, where all the plants (7B-119 and poros-200) tested were attacked by this disease. “M30” has shown a 100 % resistance against this disease, where all the plants (200) responded positively to this disease.

  The maximum increase in plant height was reported by variety Kashmir gold and Sarhad white. Kashmir gold outyielded all the hybrids under observation by producing 7768.3kg ha-1. Sarhad white produced maximum 1000 kernel weight of 370g followed by Kashmir gold with 335g. Sarhad white produced more thicher plants with 3 cobs/plant. In this screening programme Sarhad white and Kashmir gold performed well, require less time to maturity and are recommended for general cultivation in District Rawalakot, Azad Kashmir.

  Total 18 F2-monosomic lines along with the parents Poros, M30 and control plants were examined against powdery mildew which is one of the economically important wheat diseases. Screening procedure was facilitated with the division of scale 9 to 1 into resistant (5-9) and susceptible (1-4) plants. In this research, 4 chromosomes (2B, 4D, 5A and 6A) associated with the resistance against powdery mildew were identified.

  In this protocol two primers were tested against the extracted DNA, where all the amplified genotypes were found to be similar against primer AR01. The reaction of all the genotypes under observation exhibited a series of variations against primer AR02. The plants analyzed cytologically exhibited a regular mitotic behaviour except variety sargab-92, in which 2n= 40 and 41 chromosomes have been reported. All other varieties were proved to be hexaploid with 2n= 42. High molecular weight (HMW) glutenin subunits from 7 hexaploid wheat genotypes were also fractionated by SDS-PAGE, in order to characterise the material under observation. In this experiment 6 genotypes were found to be identical, while a great variation was observed within the variety sargab-92, where some extra bands have been reported.
