  Vascular Endothelial Growth Factor (VEGF) and Nitric Oxide (NO) play an important role for maintaining endothelial integrity. The purpose is to investigate the VEGF alteration during early atherosclerosis lesion formation in an animal model of hypercholesterolemia. We also measured nitrite to observe the relationship between VEGF and endothelial NO production. 20 white male rabbits randomly assigned in 2 groups (1% high-cholesterol diet, HC group, n = 14, or standard diet, control, n = 6) for 4 weeks. The serum levels of VEGF and nitrite (NO metabolite) were determined. Fatty streaks were measured in rabbit’s aortas. The results indicated that the serum level of VEGF concentration was significantly higher in hypercholesterolemic rabbits and negatively correlated with fatty streak lesions (r = -0.89, p<0.05). The serum level of nitrite was significantly higher in HC group than the control (p<0.05). There was a significant negative correlation between serum level of nitrite and VEGF (r = -0.55, p<0.05). It is concluded that, the increased VEGF in early atherosclerosis may be regarded as a safeguarding response to endothelial injury, which is responsible for maintaining endothelial integrity.

  The isolation and characterization of stem cells from an alternative tissue is a subject of intensive investigation. In the present study, we have focused on the characterization of fibroblastic cells in olfactory bulb tissue of the rat. To this end, 4-6 week old rats were killed and their olfactory bulb tissue was dissected out. Olfactory bulb derived fibroblast-like cells were recovered by adhesion to cell culture plastic. The plastic adherent cultivated cells were induced to differentiate along osteoblastic, adipogenic and chondrogenic lineages. Furthermore, the expression of some surface antigens was investigated. We obtained purified cells with spindle shaped morphology in primary culture, which differentiated into mesenchymal lineages. These cells expressed CD29 and CD90 (Thy1.1) surface antigens, but not CD31, CD34 and CD45. Our results indicate that fibroblast-like cells from the olfactory bulb are mesenchymal stem cells in nature. Taken together, our data suggest that olfactory bulb tissue may constitute a new source of mesenchymal stem cells and could be used for the treatment of injury.

  The selection of a good quality scaffold is an essential strategy for tissue engineering. Ideally, the scaffold should be a functional and structural biomimetic of the native extracellular matrix and support multiple tissue morphogenesis. However, investigators have previously shown that three-dimensional nanofibrous scaffolds are capable of influencing cellular behavior. In this study, we experimented with a three-dimensional nanofibrous scaffold fabricated from aligned-poly L-lactic acid (PLLA) for its ability to support neurogenic and hinder dopaminergic differentiation of conjunctiva mesenchymal stem cells (CJMSCs) in vitro. In this work, CJMSCs were seeded onto nanofibrous scaffolds, and were induced to differentiate along neurogenic lineages by culturing in specific differentiation media. Scanning electron microscopy imaging, RT-PCR and immunocytochemistry were used to analyze cultivated CJMSCs on scaffold and their expression of neurogenic-specific markers. We found a lack of expression of dopaminergic genes in CJMSCs seeded on align PLLA scaffold, while neurocyte-cell markers including Nestin, NSE, MAP-2 and beta-Tubulin III were expressed in these cells. On the basis of these experimental results, we conclude that the nanofibrous PLLA scaffold reported herein could be used as a potential cell carrier in neural tissue engineering and that these scaffolds could be useful for the partial inhibition of the dopaminergic differentiation of CJMSCs.
