  L-glutaminase could hydrolyze L-glutamine to L-glutamic acid which is an important ‘Umami’ substance. To obtain a salt-tolerant L-glutaminase for application in Chinese soy sauce fermentation, B. amyloliquefaciens Y-9 producing salt-tolerant L-glutaminase was isolated from mangrove sediment. The production of L-glutaminase in Solid State Fermentation (SSF) using agro-industrial residues was optimized using a central composite design of the Response Surface Methodology and the enzyme was purified which was used for further characterization including the optimum and stability of pH and temperature, effect of metal ions, substrate specificity and application to soy sauce fermentation. Under optimized conditions the experimental maximum yield of L-glutaminase reached 19.64±0.63 U gds-1 which is the highest yield obtained in SSF so far. The L-glutaminase was purified to homogeneity with final specific activity of 196.2 U mg-1 protein. The enzyme showed high activity (68% of the original activity) in the presence of 20% NaCl. The enzyme was most stable at pH 5.0 and was highly stable over an acidic pH range (3.0~7.0). These results indicate that this enzyme has a higher salt tolerance and an acidic stability. Furthermore, in Chinese soy sauce fermentation model reaction, the addition of glutaminase from B. amyloliquefaciens Y-9 was highly effective for the production of glutamic acid. This study indicates a possibility to establish economical large-scale production of L-glutaminase and this enzyme is suitable for application in soy sauce liquid fermentation with high-salt process.

  A novel γ-glutamyltranspeptidases (GGT) for the synthesis of poly-γ-glutamic acid (γ-PGA) is presented. Soil samples from the mangrove sediment were collected. One strain designated L-12 was identified whose cell extract could convert glutamine into γ-PGA. The 16S rDNA gene analysis of L-12 revealed its phylogenetic relatedness to Bacillus cereus. The GGT produced by L-12 was purified 20.3-fold. The optimum pH and temperature for γ-PGA synthesis by this GGT were 7.5 and 20°C, respectively. The results indicated that L-12 could be used as a novel bacterial GGT source for synthesizing γ-PGA production. To our knowledge, this is the first report for GGT to γ-PGA biosynthesis.

  Graphical User Interfaces (GUI) software has characteristics different from traditional software. The oracle for GUI software testing must validate the correctness of the GUI. An automated oracle based on multi-weighted Neural Networks (NN) is proposed in this paper to validate the GUI from users` viewpoint. In this approach the multi-weighted NN is used to learn the topological information in the feature space for the expected images of the graphical interface. The topological information is then used to verify the correctness of the GUI. By this method, trivial difference in the graphical interfaces can be ignored and GUI be automatically tested in the manner of human being. Experimental results show the method is of potential application in automated GUI testing.

  Oracle is one of the most difficult and expensive parts in automated software testing. It is explored in this paper to use Radial Basis Function Neural Networks (RBF NN) to construct an automated oracle model. The automated oracle generate the approximate outputs that are close to expected outputs after training. Actual outputs are then compared with the approximate outputs to determine if there is a failure when software is running. Oracle can therefore be implemented automatically and the precision be adjusted by parameters. It will save a lot of time and cost in software testing.
