  Background and Objective: Ilex cornuta (I. cornuta) is a medicinal plant that contains triterpenoid compounds as its pharmacologically active ingredients. Squalene epoxidase (SE) is a key enzyme in the triterpenoid biosynthesis pathway. The current study aimed to characterize a SE gene from I. cornuta. Methodology: IcSE2 was isolated from I. cornuta. It was predicted the secondary and tertiary structures of the IcSE2 protein, performed multiple sequence alignments and generated a phylogenetic tree. The expression level of IcSE2 was examined via Quantitative Real Time-PCR (qRT-PCR). Results: The cDNA sequence of IcSE2 was 2058 bp, with an open reading frame of approximately 1605 bp that codes for 534 amino acids. The predicted theoretical isoelectric point and molecular weight are 8.30 and 58.6 kDa, respectively. RxR, Flavin Adenine Dinucleotide (FAD) and Nicotinamide Adenine Dinucleotide Phosphate (NADPH) domains could be found in the deduced IcSE2 protein. Phylogenetic analysis showed that IcSE2 is closely related with the SEs from Araliaceae plants. IcSE2 expression level was the highest in roots, followed by leaves and male flowers and lowest in stems. Conclusion: The IcSE2 gene from I. cornuta was cloned and characterized for the first time. IcSE2 was strongly expressed in roots and leaves. These results may lay the foundation for studying the molecular regulatory mechanism of triterpenoid saponins in I. cornuta.

  Graphical User Interfaces (GUI) software has characteristics different from traditional software. The oracle for GUI software testing must validate the correctness of the GUI. An automated oracle based on multi-weighted Neural Networks (NN) is proposed in this paper to validate the GUI from users` viewpoint. In this approach the multi-weighted NN is used to learn the topological information in the feature space for the expected images of the graphical interface. The topological information is then used to verify the correctness of the GUI. By this method, trivial difference in the graphical interfaces can be ignored and GUI be automatically tested in the manner of human being. Experimental results show the method is of potential application in automated GUI testing.

  A hybrid adaptive SAGA based on mutative scale chaos optimization strategy (CASAGA) is proposed to solve the slow convergence, incident getting into local optimum characteristics of the Standard Genetic Algorithm (SGA). The algorithm combined the parallel searching structure of Genetic Algorithm (GA) with the probabilistic jumping property of Simulated Annealing (SA), also used adaptive crossover and mutation operators. The mutative scale Chaos optimization strategy was used to accelerate the optimum seeking. Compared with SGA and MSCGA on some complex function optimization and several TSP combination optimization problems, the CASAGA improved the global convergence ability and enhanced the capability of breaking away from local optimal solution.

  MicroRNAs (miRNAs) are non-coding RNAs that negatively regulate gene expression. Compelling evidences have demonstrated the important roles of miRNAs in muscle proliferation, differentiation and development processes. Cardiac muscle and skeletal muscle (longissimus dorsi muscle and psoas major muscle) are both striated muscles which perform different functions in vivo. However, little is known about the differences of miRNA expression pattern between these distinct muscles. In this study, researchers applied a deep sequencing approach to identify the miRNA transcriptomes between cardiac and skeletal muscles. Three sequencing libraries were constructed from three 210 days old pigs, a total of 56 M raw data was generated with 95.58% of them were mappable sequences. In total, 321 unique miRNAs were identified in three sequencing libraries of which 109 were differentially expressed. Ranking analysis showed that six out of the ten most highly expressed miRNAs in cardiac muscle were up-regulated compared with longissimus dorsi muscle and psoas major muscle. GO and KEGG pathway analyses for the predicted target genes of the six up-regulated miRNAs showed that the miRNAs highly expressed in CM are mainly involved in insulin and calcium signaling pathways and the constructed miRNA-mRNA interaction networks highlighted the important differences between cardiac and skeletal muscles, all these results may promote the researches on muscle-related diseases in a certain degree.

  Ultrasound measure is an important tool to evaluate carcass merit in live swine and the accuracy of ultrasonic measurement directly related to the breeding process. Here, researchers studied the effects of machine, posture and technician factors on the accuracy of ultrasonic measurement in live swine. The results indicated that the accuracy of A-mode ultrasonic machine measurement was lower than B-mode ultrasonic machine measurement and the accuracy of B-mode ultrasonic machine with equipped 3.5 MHz linear array probe performed the highest accuracy. Measured through the ultrasonic machine back fat thickness obtained from both the posture of bowing back (p<0.01) and standing posture with mouth contacting with the ground (p<0.05) were statistically significant different from the real carcass backfat thickness. Obtained from the posture of pigs were standing naturally both backfat thickness and logissimus muscle area were most approximated to the true value. During the operation of ultrasonic machine, the value of backfat thickness recognised by unexperienced technicians was statistically significantly different (p<0.01) from the true value. Thus in the process of measure both back fat thickness and logissimus muscle area through ultrasonic machine, the types of machine, the posture of pig standing and the skill level of technicians were validated to influence the obtaining of true value. In conclusion, the results indicated that the accuracy of ultrasonic measurement can be improved by suitable machines and probes, obtain picture while pig are standing naturally and skilled technicians.
