  Thresholds for extinction and persistence are important for assessing the risk of mortality in systems exposed to toxicant. In this paper, three single-species models with random perturbation in a polluted environment are proposed and investigated. One is the generalized logistic model and the other two are the stochastic resource-consumer models of Leslie and Gallopin. For each model, the survival threshold is obtained in some cases. In general, each threshold is determined by intensity of the random noise, the mean stress measure in organisms, the population intrinsic growth rate and the stress response rate.

  Due to inappropriate planning and management, accelerated urban growth and tremendous loss in land, especially cropland, have become a great challenge for sustainable urban development in China, especially in developed urban area in the coastal regions; therefore, there is an urgent need to effectively detect and monitor the land use changes and provide accurate and timely information for planning and management. In this study a method combining principal component analysis (PCA) of multisensor satellite images from SPOT (systeme pour l`observation de la terre or earth observation satellite)-5 multispectral (XS) and Landsat-7 enhanced thematic mapper (ETM) panchromatic (PAN) data, and supervised classification was used to detect and analyze the dynamics of land use changes in the city proper of Hangzhou. The overall accuracy of the land use change detection was 90.67% and Kappa index was 0.89. The results indicated that there was a considerable land use change (10.03% of the total area) in the study area from 2001 to 2003, with three major types of land use conversions: from cropland into built-up land, construction site, and water area (fish pond). Changes from orchard land into built-up land were also detected. The method described in this study is feasible and useful for detecting rapid land use change in the urban area.

  To develop viscosity-sensitive fluorescent probes, five different substituted 2-phenylbenzo[g]quinoxaline derivatives (3ae) were designed and synthesized by using benzo[g]quinoxaline as a fluorophore and phenyl ring bearing a rotatable single bond as a viscosity-sensitive unit. The fluorescence properties of these compounds were investigated in the media of the ethylene glycol–glycerol mixture with varied viscosity. It is found that 2-(4-hydroxyphenyl)benzo[g]quinoxaline (3d) and 2-(4-dimethylaminophenyl)-benzo[g]quinoxaline (3e) carrying stronger electron-donating groups in the phenyl ring show more sensitive fluorescence response to viscosity, revealing their potential use in viscosity detection and the key role of the substituted groups. The effects of solvent polarity and pH on the fluorescent properties of 3d and 3e were also discussed. The present study might be useful in developing viscosity-sensitive fluorescent probes.

  Dickkopf (Dkk) proteins are antagonists of the canonical Wnt signaling pathway and are crucial for embryonic cell fate and bone formation. Wnt antagonism of Dkk requires the binding of the C-terminal cysteine-rich domain of Dkk to the Wnt coreceptor, LRP5/6. However, the structural basis of the interaction between Dkk and low density lipoprotein receptor-related protein (LRP) 5/6 is unknown. In this study, we examined the structure of the Dkk functional domain and elucidated its interactions with LRP5/6. Using NMR spectroscopy, we determined the solution structure of the C-terminal cysteine-rich domain of mouse Dkk2 (Dkk2C). Then, guided by mutagenesis studies, we docked Dkk2C to the YWTD β-propeller domains of LRP5/6 and showed that the ligand binding site of the third LRP5/6 β-propeller domain matches Dkk2C best, suggesting that this domain binds to Dkk2C with higher affinity. Such differential binding affinity is likely to play an essential role in Dkk function in the canonical Wnt pathway.

  Wnt signaling is involved in a wide range of developmental, physiological, and pathophysiological processes and is negatively regulated by Dickkopf1 (Dkk1). Dkk1 has been shown to bind to two transmembrane proteins, the low density lipoprotein receptor-related proteins (LRP) 5/6 and Kremen. Here, we show that Dkk1 residues Arg197, Ser198, and Lys232 are specifically involved in its binding to Kremen rather than to LRP6. These residues are localized at a surface that is at the opposite side of the LRP6-binding surface based on a three-dimensional structure of Dkk1 deduced from that of Dkk2. We were surprised to find that the Dkk1 mutants carrying a mutation at Arg197, Ser198, or Lys232, the key Kremen-binding residues, could antagonize Wnt signaling as well as the wild-type Dkk1. These mutations only affected their ability to antagonize Wnt signaling when both LRP6 and Kremen were coexpressed. These results suggest that Kremen may not be essential for Dkk1-mediated Wnt antagonism and that Kremen may only play a role when cells express a high level of LRP5/6.
