  Single-strand conformation polymorphism (SSCP) analysis of ribosomal DNA (rDNA) was investigated for rapid differentiation of phenotypically similar yeast species. Sensitive tests indicated that some yeast strains with one, most strains with two, and all strains with three or more nucleotide differences in the internal transcribed spacer 1 (ITS1) or ITS2 region could be distinguished by PCR SSCP analysis. The discriminative power of SSCP in yeast species differentiation was demonstrated by comparative studies of representative groups of yeast species from ascomycetes and basidiomycetes, including Saccharomyces species, medically important Candida species, and phylloplane basidiomycetous yeast species. Though the species within each group selected are closely related and have relatively similar rDNA sequences, they were clearly differentiated by PCR-SSCP analysis of the ITS1 region, given the amplified fragments were less than 350 bp in sizes. By using SSCP analysis for rapid screening of yeast strains with different rDNA sequences, species diversity existing in a large collection of yeast strains from natural sources was effectively and thoroughly investigated with substantially reduced time and cost in subsequent DNA sequencing.

  MUSTN1 (Mustang, Musculoskeletal Temporally Activated Novel-1) gene is a musculoskeletal temporally activated novel gene which plays a key role in regulating the muscle development. In this study, the Single Nucleotide Polymorphisms (SNPs) in MUSTN1 were scanned, the association of SNPs with carcass traits in chickens were analyzed by Polymerase Chain Reaction-single-strand Conformation Polymorphism (PCR-SSCP) and DNA sequencing methods. Two variations (2120T>C and g.2160A>G) were identified when screened its entire exons and partial introns in 638 chickens. The genotypes of g.2120T>C (Mutations in exon 2) were significantly associated with breast muscle weight, muscle fiber diameters (p<0.05). The genotypes of g.2160A>G (Mutations in exon 2) were significantly associated with breast muscle weight, breast muscle weight percentage (p<0.05) and great significantly associated with muscle fiber diameters (p<0.01). MUSTN1 may act as a candidate gene of quantitative trait loci in regulating muscle growth.

  MicroRNAs (miRNAs) are small, non-protein-coding RNAs that function as tumour suppressors or oncogenes. A single nucleotide change in the sequence of pre-miRNA can affect miRNA expression, so single-nucleotide polymorphisms (SNPs) in pre-miRNA may be biomarkers for biomedical applications. In this study, we performed a genetic association study between the SNP (rs11614913) in pre-miRNA-196a and esophageal squamous cell carcinoma (ESCC) susceptibility in a case–control study. We found that the homozygote CC of this SNP increased the risk of ESCC compared with the homozygote TT and the risk was more evident among smokers than non-smokers. Therefore, this functional SNP may be a biomarker for ESCC outcome.

  A method to authenticate the integrity of audio aggregation is proposed in this study. The vector-sharing scheme is used to fragile watermarking technology because it is very sensitive when recovering the secret. In this algorithm, the key sharing ideology is introduced into audio aggregation watermarking algorithm by separating the watermark in different audio works of the audio aggregation. When detected the original watermark can be recovered without original audio aggregation. The experimental results show that the proposed algorithm has strong vulnerability, good imperceptibility and can locate the audio work (s), which suffered attack in the audio aggregation.

  Steganography techniques can be used to embed secret information into audio signals. A Huffman coding section-based steganographic scheme for MPEG-2/4 Advanced Audio Coding (AAC) audio is proposed in this study. Based on the characteristics of Huffman coding section, the scheme hides secret information by modifying the sections of Huffman coding without affecting its normal process of encoding. Experimental results are given to show that the proposed scheme not only has a larger capacity, 15 bits per frame averagely but also does not change the statistical properties of the audio carrier which means having certain undetectability. In addition, the scheme does not affect the quality of audio carrier, having good imperceptibility.

  As a traditional medicinal herb in China, Penthorum chinense Push from Gulin county is regarded as genuine regional drug with better clinical effects. To discriminate the geographical origin of six P. chinense samples cultivated in three provinces, Randomly Amplified Polymorphic DNA (RAPD) analysis was carried out with an improved method to increase the resolution and production using 10 mer-random primers. Similarity index was ranged from 0.61 to 0.97, which demonstrated that samples from different localities displayed similar band patterns. However, based on the analysis of selected 13 primers, primers SBS-I9, SBS-I20 and SBS-Q9 produced distinguishable bands among Sichuan, Yunnan and Hubei P. chinense. T-test of mean S.I. values of six accessions demonstrated the significant differences between Luzhou or Sichuan samples and others. Cluster analysis indicated that cultivars with close geographic distributions were clustered together consequently. This suggested that there are RAPD site variations and local specialized genotypes among the six samples. The results indicated that RAPD analysis is effective in distinguishing the geographical origin of genuine regional drug P. chinense grown in Luhzou, Sichuan. The approach is a valuable tool to authenticate other morphologically similar herbal medicinal materials.

  The photosynthetic properties and photosynthetic responses to drought stress in the pot experiment of three chicory (Cichorium intybus L.) strains with different drought resistance, space-mutagenesis-bred strains PA-82 and PA-43 and their initial strain PA-57 were studied using Li-6400 portable photosynthesis system. The results showed that diurnal time courses of net Photosynthetic rate (Pn), Transpiration rate (Tr), stomatal Conductance (Cond) presented as double-peak curves. There was an obvious midday Pn depression, which was caused by nonstomatal factors. Highest Pn, Tr and Cond values were observed in PA-82 and lowest corresponding values in PA-43 (p<0.05). Intercellular CO2 Concentration (Ci) of three strains showed an opposite course to that of Pn which reached its valley at 10:00-12:00. PA-82 had the highest Water Use Efficiency (WUE), followed by PA-57, then PA-43. Light Saturation Points (LSP) of three strains were between 1207-1264 μmol./m2/sec and Light Compensation Points (LCP) were between 14.55-14.85 μmol/.m2/sec, both indicating good adaptation to changes of light intensity. Pn, Tr, Cond, maximal photochemical efficiency of PS II in dark adaptation (Fv/Fm), potential photochemical efficiency (Fv/F0) and coefficient of Photochemical quenching (qP) decreased with the aggravation of water deficit. Among the three strains, the parameters of PA-43 dropped to a greater extent at a highest speed, followed by PA-57 then PA-82. With the aggravation of water deficit, Ci and co-efficient of nonphotochemical quenching (qN) increased to a greater extent at a highest speed in PA-43. The results suggest that PA-82 and PA-57 have higher resistance against severe water deficit in comparison with PA-43.

  A 3d-4f heterometallic coordination polymer, lcub[EuCu(C2O4)(na)2] · 2H2Orcubn (1) [Hna=nicotinic acid], has been synthesized by hydrothermal reaction of lanthanide oxides, Cu(I), and pyridine-2,3-dicarboxylic acid. 1 features a 3-D pillar-layered coordination framework constructed from two-dimensional lanthanide-carboxylate layers and Cu(na) pillars. Interestingly, the na ligand was obtained by the in situ decarboxylation of pyridine-2,3-dicarboxylic acid.

  Na,K-ATPase and inositol 1,4,5-trisphosphate (IP3) receptor (IP3R) can form a signaling microdomain that in the presence of ouabain triggers highly regular calcium oscillations. Downstream effects include NF-κB activation. Here we report that ankyrin B (Ank-B), expressed in most mammalian cells, plays a pivotal role in the function of the Na,K-ATPase/IP3R signaling microdomain. In studies performed on a monkey kidney cell line, we show that Ank-B co-precipitates with both Na,K-ATPase and IP3R. We identify the N terminus tail of the Na,K-ATPase catalytic subunit and the N-terminal portion 1-604 of the IP3R as novel binding sites for Ank-B. Knockdown of Ank-B with small interfering RNA reduced the expression of Ank-B to 15-30%. This down-regulation of Ank-B attenuated the interaction between Na,K-ATPase and IP3R, reduced the number of cells responding to pM doses of ouabain with calcium oscillations, altered the calcium oscillatory pattern, and abolished the ouabain effect on NF-κB. In contrast, Ank-B down-regulation had no effect on the ion transporting function of Na,K-ATPase and no effect on the distribution and apparent mobility of Na,K-ATPase in the plasma membrane.
