  A sensitive method for specific detection of viable Escherichia coli O157:H7 cells, including viable but nonculturable (VBNC) cells, in water samples was developed. This method involved capture of the bacterial cells on a low-protein-binding membrane and direct extraction and purification of RNA followed by reverse transcription-PCR and electronic microarray detection of the rfbE and fliC genes of E. coli O157:H7. It detected as few as 1 CFU of E. coli O157:H7 in diluted cultures, 3 to 4 CFU/liter in tap water, 7 CFU/liter in river water, and 50 VBNC cells in 1 liter of river water, demonstrating the best limit of detection reported to date for VBNC cells in environmental water samples.

  Ca2+ is an important intracellular messenger during oocyte development. Ca2+ oscillations are dependent upon the Endoplasmic Reticulum (ER), an internal Ca2+ store capable of releasing and re-sequestering Ca2+. However, mechanisms that regulate oocyte fragmentation are unclear. The aim of the current study was to investigate the mechanism of Ca2+ release and ER cluster distributions during fragmentation of aging culture porcine oocytes. After oocytes were cultured over a 40 h interval, ER cluster distributor and Ca2+ amplitudes significantly decreased. Oocyte fragmentation appeared after cells had been cultured 24 h and reached its peak at 48 h. At 40 h, normal morphology oocytes (NMor ) had low Ca2+ amplitude and little ER cluster distributor; partial fragmentation (PFra) oocytes had less frequent and shorter amplitudes of Ca2+ release with ER cluster distribution partly aggregated in cytoplasm; whereas complete fragmentation oocytes (Cfra), which were increased by low voltage EA or enucleation, had no Ca2+ amplitude or ER cluster distribution. The number of PFra oocytes that proceeded to CFra following aging was increased in a Ca2+ free medium. We therefore conclude that extracellular Ca2+ abnormality resulted in altered intracellular Ca2+, leading to changes in oocyte fragmentation dynamics.

  Endotoxin removal from recombinant proteins is considered to be an indispensable step in bioprocesses before the application in biomaterials for pharmaceutical industry. This study was carried out using nonionic detergent Triton X-114 to eliminate endotoxin molecules from human-like collagen preparations. With the evaluation of four factors (Triton X-114 concentration, temperature, time and pH) which were mainly affecting the two-phase extraction behavior, the optimal experimental condition was determined. Under this condition, nearly 98~99% endotoxins were removed with high protein recovery (95.1%). The residual Triton X-114 was efficiently separated from HLC preparation via ultrafiltration procedure with microgram amounts of residues left. Through toxicity test in vitro, the processed HLC was proved to be non-toxic to BHK21 and acceptable as a novel biomaterial. Therefore, Triton X-114 two-phase extraction can be used as an effective way to remove endotoxins from HLC preparations.

  Preschool-aged children are aware of differences in the race and abilities of the people around them. Given this awareness it is important to promote children’ s acceptance of diversity in the preschool period. The goals of this study were to assess the extent to which child care centres provide diversity instruction through classroom activities, materials and displays. The extent to which structural quality characteristics (e.g. staff training and education) contribute to diversity-positive classrooms was also examined. Data were collected from 103 preschool classrooms in 64 child care centres serving a population of ethnically diverse families in Toronto, Canada. On average, these classrooms were found to be diversity-positive environments. Hierarchical linear model analyses indicate that utilising a variety of teaching formats, higher salaries, greater supervision and having higher proportions of children who receive a child care subsidy predicted higher scores on a diversity instruction and materials index. This index was largely based on classroom observations. In contrast, lower levels of education and salary predicted staff reports of diversity-promoting activities. These latter counter-intuitive results are interpreted in light of potential self-presentation biases.

  The study analyzed the current popular unstructured super-peer topology and the search technique. In order to avoid the high expense of topology construction, the huge cost of topology repair and the low efficiency of search, a kind of novel construction method of orderly layered super-peer topology guided by the safety peer (OLST) is presented and the search function is realized in this study. After the leaf peers being transferred and layered, the new coming peer will be joined by an assigned safety super-peer. An idea of among friends with similar interests is used to improve the search efficiency of the topology. The experimental results indicate that the method can effectively balance the super-peer load and significantly decrease the construction and repair cost of the topology. Furthermore, the search efficiency is also increased.

  Digital community involves e-government, e-commerce, smart health and other applications. With the increase of customers and types of business, it becomes more important for digital community to process massive data efficiently. Although, the current cloud-based applications can provide some elastic and on-demand calculation abilities to digital community, their underlying programming models still have certain limitations. This study aims to provide a new framework of massive data processing for digital community. In the framework, multiple programming models are adopted and each programming model handles the specific calculations that they do best. These calculations mainly include embarrassingly parallel calculation, iteration calculation and data-dependent complex calculation. To improve the performance of the framework, the programming model connection pool and the virtual subnet are designed and applied. Compared to Hadoop and its modified version, on average, the proposed framework can reduce runtime by 1.32. The experimental results show that the proposed framework has higher generality and efficiency. Moreover, it is reasonable and valuable for digital community to analyze comprehensively trade area on geographical location and business volume.

  With the continuous development of society and the prevalence of computer and network technology. How to ensure the security of information in the course of transmission have become the most important things for people at present. With this background, we studied how to realize encryption and decryption of the RSA (Initials of Ron Rivest, Adi Shamirh, LenAdleman) encryption technology. This study mainly introduces the application of RSA algorithm in encryption and decryption, mentions the technology of digital signature. Also introduces in the process of implementation of RSA algorithm in Visual Studios environment and operation results. Using this system for encryption and decryption of information, theoretically, good results were obtained in safety and reliability.

  Multi-core processors are becoming ubiquitous with the continuous development of hardware technology. But many applications are sequential applications and they receive no benefits running on multi-core processors. Addressing this problem, using parallel techniques improve the sequential program running on the multi-core processors. In this paper, we introduced three parallel types of thread level parallelism. Typical DOALL, DOACROSS, DSWP and PS-DSWP techniques are described. These techniques can explore the parallel from sequential application, but much dependence is not easily predictable or manifests them infrequently by the non-speculative transformation. So many speculative techniques, such as thread level speculation (TLS), Speculation DSWP (SpecDSWP), Speculative PS-DSWP (SpecPS-DSWP) and Interprocedural SpecPS-DSWP (iSpecPS-DSWP), are proposed to break problematic dependences to enhance parallelism. We introduced these speculative parallel techniques and described their execution. SpecDSWP, SpecPS-DSWP and iSpecPS-DSWP are compared from supporting speculation types, memory version and implement steps. At last, some extended TM systems which support TLS techniques are analyzed from thread spawning mechanism, context passing mechanism and sequential ordering.

  RFID-based location awareness is becoming the most important issue in many fields in recent years, such as ubiquitous computing, mobile computing. However, the RF system is noise-limited, which leads to the readers can’t read the information from tags timely and accurately, especially for RFID localization network. This study proposed a novel RFID indoor localization method based on Received Signal Strength Indicator (RSSI) and Packet Received Ratio (PRR). To do so, the environmental factors affecting the link quality are analyzed and the location awareness data is collected by RFID equipment using non-coherent Frequency Shift Keying (FSK) as modulation scheme and Not Return to Zero (NRZ) as encoding scheme. Then the relationship model between RSSI, PRR and distance is established based on the classic radio propagation model in localization field and theoretical analysis of the normal probability distribution of PRR is conducted. The experimental results show that our approach is valid and proper and also contributes to a novel perspective and theoretical support on the further study and application of RFID indoor localization.

  In recent years, with the development of data warehouse and Web technology, more and more attention has been paid to multiple applications of the data in warehouse. However, data quality issue is one of the biggest obstacles to the success using of data warehouse project for many enterprises. So a data audit model is proposed and the relevant methods are studied in the study based on the characteristics of the telecommunications industry. Further, a three data layer audit method, consisted of audit mode data file level, record level and index level, is constructed during Extraction Transformation Loading (ETL) process. It can effectively improve the data quality of data warehouse.

  Wireless Radio Frequency (RF) technology, which can take the place of cable, has been widely used in many current fields as one of information technologies with widest horizon of development. The problems caused by the environment restrictions and the unfavorable conditions for cable wiring have been solved because of the emergence of the wireless. Its three important characteristics and advantages are low costing, low power consumption and peer-to-peer communication. During the design procedure, the system is divided into three major modules according to the functional structure, namely central control module, wireless transceiver module and computer dealing software, The central control module uses Sygnal company's low power enhanced 51 MCU C8051F310 as CPU, mainly to complete the data acquisition and processing. The wireless transceiver module is made up of TI CC1101 wireless transceiver chip, which mainly realizes wireless data transmission and The computer dealing software real-timely displays and deals with the data through visual, friendly programming interface, based on C#.NET. Final achievement of the system design is two compact communication devices, one of them connects with PC through RS-232 interface, System is operated through the graphical user interface based on windows, which is very convenient to operate. Debugging result indicated that data acquisition, processing and transmission could be achieved in wireless or wired way under the control of PC and it visually displays the results to the user, satisfies request of the wireless power monitoring data communication system, reaches the technical index.

  Background and Objective: The laparoscopic technique has been widely used in clinical gynaecology because of its advantages on smaller wounds, low perioperative stress responses and fewer postoperative complications. It is necessary to actively examine a drug with accurate effect and high safety to reduce organ damages and promote smooth operation. This study aimed to explore the perioperative effects of hydromorphone on oxidative stress, renal and immune functions and inflammatory factors in gynaecological laparoscopic surgery under general anaesthesia. Materials and Methods: Sixty patients who underwent gynaecological laparoscopic surgery under general anaesthesia were selected and divided into 2 groups by random number table method, including a control group (n = 30, treated with the same amount of normal saline) and an observation group (n = 30, pretreated with hydromorphone). Results: The observation group had shorter anaesthesia onset time, block time and postoperative recovery time and lower anaesthesia dosage than the control group (p<0.05). The observation group had lower serum O2, H2O2, OH, MDA content, BUN, UA, Cr content and TNF-α, IL-6 and S-100 β contents at T1 and T2 but had higher GSH-Px and SOD content and serum Terg, TGF-β1, IL-10 levels and serum CD3+ and CD4+ levels at T1 and T2 than the control group (p<0.05). Conclusion: Pretreatment with hydromorphone for patients undergoing gynaecological laparoscopic surgery under general anaesthesia can relieve cellular immune suppression induced by surgical stress, improve oxidative stress responses and renal function, reduce the expression of inflammatory factors and relieve the pain degree of patients.

  Two new divalent metal aminodiphosphonates with a layered structure, Mn2[(HL)(H2O)F]·H2O (1) and Cd3.5[(HL)L] (2) (H4L = CH3CH2CH2N(CH2PO3H2)2), have been hydrothermally synthesized and characterized by single-crystal X-ray diffraction as well as with infrared spectroscopy, elemental analysis and thermogravimetric analysis. In 1, two MnO4F2 and two MnO5F polyhedra are interconnected via edge-sharing into a tetramer, and such tetramers are bridged by the diphosphonate ligands into a Mn(II) phosphonate layer in the ab-plane. The structure of 2 also features a 2D layered structure, in which the CdO5N and CdO6 polyhedra are interconnected into a 1D chain. The chains are then cross-linked via phosphonate oxygen atoms to form Cd(II) phosphonate layers in the ab-plane.

  Solution reactions of silver(I), copper(I), cadmium(II) and zinc(II) salts with 1,3-imidazolidine-2-thione (imdt) under diverse conditions yielded four complexes: [Cd(SC3H6N2)2(Ac)2] (1), [Zn(SC3H6N2)2(Ac)2] (2), [Cu2(SC3H6N2)6]SO4 (3) and [Ag2(SC3H6N2)6]SO4 (4). Complexes 1 and 2 are 1D and 2D hydrogen-bond aggregations. Complexes 3 and 4 are isostructural 3D hydrogen-bond networks. The diverse coordination modes of imdt and different anions are the major factors for three distinct hydrogen-bond structures.

  The potential of using small RNA molecules as therapeutic agents has been extensively explored, antisense RNA, ribozyme, aptamer, decoy and more recently siRNA have been demonstrated to be highly efficient in inhibiting a number of pathogenic viruses including human immunodeficiency virus, hepatitis B and C virus, poliovirus and influenza virus. The specificity and potency of the sequence-specific agents such as antisense, ribozyme and siRNA in particular, imply that these strategies will prove to be promising therapeutics for treating viral infections, although the antiviral efficacy may be limited by emergence of escape variants. Distinct from the reagents targeting viral RNA, decoy and aptamer inhibit viral replication by binding and thus inactivating the viral component such as regulatory gene product and viral enzyme. This review provides an up-to-date overview of the progress and problems in small RNA-based antiviral approaches, with a focus on their therapeutic utility, delivery and unwanted side effects.

  The concentrative nucleoside transporter (CNT) protein family in humans is represented by three members, hCNT1, hCNT2, and hCNT3. Belonging to a CNT subfamily phylogenetically distinct from hCNT1/2, hCNT3 mediates transport of a broad range of purine and pyrimidine nucleosides and nucleoside drugs, whereas hCNT1 and hCNT2 are pyrimidine and purine nucleoside-selective, respectively. All three hCNTs are Na+-coupled. Unlike hCNT1/2, however, hCNT3 is also capable of H+-mediated nucleoside cotransport. Using site-directed mutagenesis in combination with heterologous expression in Xenopus oocytes, we have identified a C-terminal intramembranous cysteine residue of hCNT3 (Cys-561) that reversibly binds the hydrophilic thiol-reactive reagent p-chloromercuribenzene sulfonate (PCMBS). Access of this membrane-impermeant probe to Cys-561, as determined by inhibition of hCNT3 transport activity, required H+, but not Na+, and was blocked by extracellular uridine. Although this cysteine residue is also present in hCNT1 and hCNT2, neither transporter was affected by PCMBS. We conclude that Cys-561 is located in the translocation pore in a mobile region within or closely adjacent to the nucleoside binding pocket and that access of PCMBS to this residue reports a specific H+-induced conformational state of the protein.

  The 55-kDa TNFR1 (type I tumor necrosis factor receptor) can be released to the extracellular space by two mechanisms, the proteolytic cleavage and shedding of soluble receptor ectodomains and the release of full-length receptors within exosome-like vesicles. We have shown that the brefeldin A-inhibited guanine nucleotide exchange protein BIG2 associates with TNFR1 and selectively modulates the release of TNFR1 exosome-like vesicles via an ARF1- and ARF3-dependent mechanism. Here, we assessed the role of BIG2 A kinase-anchoring protein (AKAP) domains in the regulation of TNFR1 exosome-like vesicle release from human vascular endothelial cells. We show that 8-bromo-cyclic AMP induced the release of full-length, 55-kDa TNFR1 within exosome-like vesicles via a protein kinase A (PKA)-dependent mechanism. Using RNA interference to decrease specifically the levels of individual PKA regulatory subunits, we demonstrate that RIIβ modulates both the constitutive and cAMP-induced release of TNFR1 exosome-like vesicles. Consistent with its AKAP function, BIG2 was required for the cAMP-induced PKA-dependent release of TNFR1 exosome-like vesicles via a mechanism that involved the binding of RIIβ to BIG2 AKAP domains B and C. We conclude that both the constitutive and cAMP-induced release of TNFR1 exosome-like vesicles occur via PKA-dependent pathways that are regulated by the anchoring of RIIβ to BIG2 via AKAP domains B and C. Thus, BIG2 regulates TNFR1 exosome-like vesicle release by two distinct mechanisms, as a guanine nucleotide exchange protein that activates class I ADP-ribosylation factors and as an AKAP for RIIβ that localizes PKA signaling within cellular TNFR1 trafficking pathways.

  In humans, the SLC28 concentrative nucleoside transporter (CNT) protein family is represented by three Na+-coupled members; human CNT1 (hCNT1) and hCNT2 are pyrimidine and purine nucleoside-selective, respectively, whereas hCNT3 transports both purine and pyrimidine nucleosides and nucleoside drugs. Belonging to a phylogenetic CNT subfamily distinct from hCNT1/2, hCNT3 also mediates H+/nucleoside cotransport. Using heterologous expression in Xenopus oocytes, we have characterized a cysteineless version of hCNT3 (hCNT3C-). Processed normally to the cell surface, hCNT3C-exhibited hCNT3-like transport properties, but displayed a decrease in apparent affinity specific for Na+ and not H+. Site-directed mutagenesis experiments in wild-type and hCNT3C-backgrounds identified intramembranous Cys-561 as the residue responsible for this altered Na+-binding phenotype. Alanine at this position restored Na+ binding affinity, whereas substitution with larger neutral amino acids (threonine, valine, and isoleucine) abolished hCNT3 H+-dependent nucleoside transport activity. Independent of these findings, we have established that Cys-561 is located in a mobile region of the hCNT3 translocation pore adjacent to the nucleoside binding pocket and that access of p-chloromercuribenzene sulfonate to this residue reports a specific H+-induced conformational state of the protein (Slugoski, M.
