  Genetic variation at 20 microsatellite loci and genetic diversity were examined for Yangzhou Chicken. Observed and effective number of alleles across the microsatellite loci varied from 2-6 with an overall mean of 3.778 and 2.404, respectively. Observed and effective heterozygosity varied from 0.129-0.755 with an average of 0.422 and 0.517, respectively. Average polymorphism information content was 0.464. The genetic structure indicated that Yangzhou Chickens have substantial genetic variation. Population showed fairly high level of inbreeding (Fis = 0.184) and global heterozygote deficit. The allele frequency distribution is displayed an L shape, suggesting that no recent bottleneck affecting the genetic variability occurred. The information generated in this study will greatly aid in the establishment of effective breeding strategies for Yangzhou Chicken and may further be utilized for studying differentiation and relationships among different chicken breeds.

  Lysozyme plays important role in vertebrates and invertebrates for its unique protein structure and enzyme activity. In the present study, the potential physiological function of the chicken lysozyme gene (LYZ) was studied by SNP. 329 J+ line Jinghai Yellow chicken were involved in this experiment. To determine gene mutations, DNA analysis of the LYZ gene, including Single Strand Confirmation Polymorphism (SSCP) and direct DNA sequencing, was performed. Chicken Body Weight (BW) were analyzed by GLM of SAS software. DNA sequencing demonstrated a G111A transition of exon 1, a T1426C transition and a C1492T transition of exon 2, all of them were silent mutations that caused no alteration in amino acid sequence. Statistical analysis indicated that, AA and CC genotype related with high BW, TT and CN genotype contributed to low BW. Therefore, results suggested that there was a possibility of the LYZ gene SNPs acting as a genetic marker for growth traits of Jinghai Yellow chicken.

  Xiaoshan chicken is a well-known breed in China. It has good meat quality and three yellow character. SR92A chicken and Xiaoshan chicken are parents. Sixty five offsprings were reared. DNA Fingerprint (DFP) of parental strains were conducted using one middle-repetitive DNA probe EAV (endogenous avian retroviral). Based on presence or absence of band J (3.48 kbp) in their parents, we carry out 4 mating combinations. -/- for absence of band J in both parents; -/+ for absence in sire, presence in dam; +/- for presence in sire, absence in dam; +/+ for presence in both parents. Quantitative traits recorded for offspring are body weight at 75 days and slaughter traits. The results show that the 75 days weight heterosis generated by -/- type is 74.1 g higher than generated by non-selected populations. We can infer that the allele may be corresponding with the different expression of genes between parents and offsprings. We find band J allele has a bearing on weight heterosis, which will lay a solid foundation for locating weight heterosis QTL.
