  Quinolone resistance is an emerging problem in China. To investigate the prevalence of the plasmid-mediated quinolone resistance genes qnr and aac(6`)-Ib-cr, a total of 265 clinical isolates of Escherichia coli, Klebsiella pneumoniae, Citrobacter freundii, and Enterobacter cloacae with ciprofloxacin MICs of ≥0.25 µg/ml were screened at nine teaching hospitals in China. The qnrA, qnrB, qnrS, and aac(6`)-Ib genes were detected by PCR. The aac(6`)-Ib-cr gene was further identified by digestion with BtsCI and/or direct sequencing. The qnr gene was present in significantly smaller numbers of isolates with cefotaxime MICs of <2 µg/ml than isolates with higher MICs (≥2.0 µg/ml) (20.6% and 42.1%, respectively; P < 0.05). aac(6`)-Ib-cr was present in 17.0% of the isolates tested, and 7.9% of the isolates carried both the qnr and the aac(6`)-Ib-cr genes. Among the isolates with cefotaxime MICs of ≥2.0 µg/ml, qnr and aac(6`)-Ib-cr were present in 65.7% and 8.6% of E. cloacae isolates, respectively; 65.5% and 21.8% of K. pneumoniae isolates, respectively; 63.3% and 26.7% of C. freundii isolates, respectively; and 6.5% and 16.9% of E. coli isolates, respectively. The 20 transconjugants showed 16- to 128-fold increases in ciprofloxacin MICs, 14 showed 16- to 2,000-fold increases in cefotaxime MICs, and 5 showed 8- to 32-fold increases in cefoxitin MICs relative to those of the recipient due to the cotransmission of blaCTX-M-14, blaCTX-M-3, blaDHA-1, blaSHV-2, and blaSHV-12 with the qnr and aac(6`)-Ib-cr genes. Southern hybridization analysis showed that these genes were located on large plasmids of different sizes (53 to 193 kb). These findings indicate the high prevalence of qnr and aac(6`)-Ib-cr in members of the family Enterobacteriaceae and the widespread dissemination of multidrug resistance in China.

  By using multilocus sequence analysis, five Borrelia valaisiana-related strains isolated from rodents and ticks in southwestern China were eventually classified as a new genospecies of B. burgdorferi sensu lato rather than B. valaisiana. The finding explained the differences in transmission cycle and phenotype between B. valaisiana strains from Europe and B. valaisiana-related strains from eastern Asia.

  The synthesis and characterisation of a main-chain chiral smectic liquid crystalline thiol-ene polymer system is described. The chemical structure is designed based on the structure of W317, a mesogen with a large electroclinic effect in the Smectic A* (SmA*) phase. The novel thiol-ene monomer K0902 has a large surface electroclinic effect with an unexpected small electroclinic effect in the SmA* phase. The corresponding thiol-ene oligomer poly-K0902 is characterised by gel permeation chromatography (GPC), differential scanning calorimetry (DSC), polarised optical microscopy (POM), X-ray diffraction (XRD) and shows an expected I-SmA*-Glass phase sequence, but the electroclinic angle is relatively small.

  The use of Sudan I as an additive in food products has been prohibited in the European Union and many other countries. In this study, a highly sensitive and specific monoclonal antibody (mAb)-based indirect competitive enzyme-linked immunosorbent assay (ELISA) for the detection of Sudan I in food samples was developed. The hapten derivative with a three-carbon-atom length of carboxylic spacer at the azobound para-position was synthesized and coupled to carrier proteins. The hapten–bovine serum albumin (BSA) conjugate was used as an immunogen, while the hapten–ovalbumin (OVA) conjugate was applied as a coating antigen. The mAb against Sudan I was produced by hybridoma technique and the corresponding ELISA was characterized in terms of sensitivity, specificity, precision and accuracy. At optimal experimental conditions, the standard curve was constructed in concentrations of 0.1–100ngmL−1. The values of IC50 for nine standard curves were in the range of 1.1–2.0ngmL−1 and the LOD at a signal-to-noise ratio of 3 (S/N=3) was 0.07–0.14ngmL−1. The cross-reactivity values of the mAb with Sudan II, III and IV were 9.5%, 33.9% and 0.95%; no cross-reactivity was found with other six edible colorants: Lemon yellow, Bright blue, Indigotin, Kermes, Amarant and Sunset yellow, indicating the assay displays not only high sensitivity but also high specificity as well. The organic solvent effect on the assay was tested. It was observed that the ELISA was tolerated to 30% of methanol and 10% of acetonitrile without significant loss of IC50 value. Six food samples were spiked with Sudan I and the methanolic extracts after appropriate dilution were analyzed by ELISA. Acceptable recovery rates of 88.2–110.5% and coefficients of variation of 2.5–17.4% were obtained. The ELISA for nine spiked samples was confirmed by high-performance liquid chromatography (HPLC) with a high correlation coefficient of 0.9840 (n=9). The mAb-based ELISA proven to be a feasible quantitative/screening method for Sudan I analysis in food samples with the properties of high sensitivity, specificity, simplicity of sample pretreatment, high sample throughput and low expense.

  Exon IV of SLC2A1, a multiple facilitator superfamily (MFS) transporter gene, is particularly susceptible to mutations that cause GLUT1 deficiency syndrome, a human encephalopathy that results from decreased glucose flux through the blood-brain barrier. Genotyping of 100 patients revealed that in a third of them who harbor missense mutations in the GLUT1 transporter, transmembrane domain 4 (TM4), encoded by SLC2A1 exon IV, contains mutant residues that have the periodicity of one face of a kinked α-helix. Arg-126, located at the amino terminus of TM4, is the locus for most of the mutations followed by other arginine and glycine residues located elsewhere in the transporter but conserved among MFS proteins. The Arg-126 mutants were constructed and assayed for protein expression, targeting, and transport capacity in Xenopus oocytes. The role of charge at position 126, as well as its accessibility, was investigated in R126H by determining its activity as a function of extracellular pH. The results indicate that intracellular charges at the MFS TM2–3 and TM8–9 signature loops and flanking TMs 3, 5, and 6 are critical for the structure of GLUT1 as are TM glycines and that TM4, located at the catalytic core of MFS proteins, forms a helix that surfaces into the extracellular solution where another proton facilitates transport.

The total number of secondary structures that have given loops with minimal length m(m≥0 or m≥1) and minimal stack length l(l≥0 or l≥1) is computed, under the assumption that all base pairs can occur.

  Captive farming of alpine musk deer (Moschus sifanicus) in China has been used for conservation and harvesting of musk since the mid 1950s. Despite this long history, management practices and captive breeding have been primarily based on trial and error due to lack of behavioral and ecological information about this vulnerable species. Understanding behavioral patterns plays a vital part in determining appropriate management systems; hence the aim of this study was to determine the effect of captivity on behavioral patterns of alpine musk deer by comparing wild-caught and the captive-born alpine musk deer. From August 2002 to January 2003, the behavioral patterns of 30 wild-caught (WC) and 15 captive-bred (CB) adult alpine musk deer were recorded at Xinglongshan Musk Deer Farm (XMDF), located in Xinglongshan National Nature Reserve, Gansu province, China. Focal sampling was used to observe the frequencies of 12 behavior categories. The behavioral patterns of WC and CB musk deer were found to be similar; however, when gender was considered, male WC deer showed a significantly higher frequency of agonistic interaction. These preliminary results suggest that captivity has had no immediate impact on the behavioral patterns of captive alpine musk deer despite 10 generations of captivity. Therefore, the alpine musk deer is not suited for domestication and further investigation into the effectiveness of musk deer farming for the purpose of harvesting musk should be undertaken.
