  Cytotoxic and anti-angiogenic properties of n-hexane extract of Sandoricum koetjape stem bark were investigated in vitro. 2,3-bis-(2-methoxy-4-nitro- 5-sulfophenyl)-2H-tetrazolium- 5-carboxanilide (XTT) cell proliferation assay was used to study the cytotoxic properties on Human Umbilical Vein Endothelial Cell (HUVEC) and both colon cancer and normal cell lines; HCT-116, HT-29 and CCD-18CO. Rat aorta ring assay was used to study the anti-angiogenic properties of the extract. At 100 μg mL-1, the extract showed 94±5.5% inhibition of the outgrowth of the blood vessels from the rat aorta rings. The extract also showed a dose dependent growth inhibition of all tested cell lines, IC50 values against HCT-116, HUVEC, CCD-18CO and HT-29 were 14, 23, 50 and 52 μg mL-1, respectively. At 50 μg mL-1, the extract had potently induced apoptotic cell death of HCT-116 colon cancer cell line by inducing caspases 3 and 7 activity. These results showed that n-hexane extract of S. koetjape possess both anti-angiogenic and apoptotic properties on colon cancer cell lines making it a good candidate for further studies.

  Both cytotoxic and apoptotic properties of the stem bark extract of Sandoricum koetjape were investigated in vitro. 2,3-bis-(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium- 5-carboxanilide (XTT) cell proliferation assay was used to study the cytotoxic properties on three breast cancer cell lines and one normal cell line; MCF-7, MDA-MB-231, T47D and MCF-10A, respectively. Caspase Glo 3/7 assay was used to study the apoptotic activity on MCF-7 as a model cell line. The n-hexane extract showed a dose dependent growth inhibition of all tested cell lines with IC50 values between 44 and 48 μg mL-1. At 100 μg mL-1, the extract induced apoptotic cell death of MCF-7 by inducing activity of the effector caspases 3 and 7. These results indicated that n-hexane extract of S. koetjape has cytotoxic and apoptotic properties on breast cancer cell lines making it a good candidate for further studies.

  Angiogenesis is essential for the growth and metastasis of most solid malignancies. Accordingly, tumor angiogenesis is an important pharmacological target for cancer prevention and treatment. This study was conducted to study antiangiogenic activity of some tropical plants. Seven plants were extracted successively with n-hexane and methanol to prepare 18 extracts and antiangiogenic properties of the extracts were studied by rat aorta ring assay. Nine extracts showed more than 50% inhibition of the blood vessels outgrowth from the primary tissue explants. The MTT assay was used to study the cytotoxic activities of extracts with more than 50% inhibition. The Human Umbilical Vein Endothelial Cells was used as a test cell line versus two human colon cancer cell lines HCT-116 and HT-29, two human breast cancer cell lines MCF-7 and T47D and one hepatocarcinoma cell line HepG-2. The selectivity index on Human Umbilical Vein Endothelial Cells was calculated. The selectivity index results indicated antiangiogenic activities of three plants Parkia speciosa, Syzygium campanulatum and Sandoricum koetjape. The results presented in this research article make these plants good candidates for further studies to purify the active compounds and to study in vivo antiangiogenic activity.

  Catharanthus roseus (Apocynaceae), in Malaysia known as Kemuning cina, well known for being rich in alkaloids was investigated for its cytotoxic activity by using MTT assay against Human Colorectal Carcinoma Cell Line (HCT 116). The preliminary cytotoxicity study demonstrated dose independent cytotoxic activity of the methanol extract of C. roseus when screened against HCT-116 colorectal carcinoma cell line. n-hexane, chloroform and methanol fractions also showed dose independent cytotoxic activity with chloroform fraction showing the highest activity. Water fraction showed a minor cytotoxic activity, vindoline also showed some cytotoxic activity at 200 μg mL-1. Catharanthine showed the most promising activity while dose dependent cytotoxic activity of its IC50 value was found to be at 60 μg mL-1. Simple and facile method has been developed for the isolation of compounds catharanthine and vindoline from this plant.
