Interaction between Th9 cells, Interleukin-9 and Oxidative Stress in Chronic Lymphocytic Leukemia

Abstract: Chronic Lymphocytic Leukemia (CLL) is a cancer of B lymphocytes, originating within the bone marrow and poignant most body tissues. The T helper 9 (Th9) cell is the chronic T helper cells related to Th2 immunity and secretes IL-9 which make these cells contribute to each protecting immunity and immunopathological malady. This review focused on the role of Th9 cells and oxidative stress in cancer immunity especially in CLL. Also, it summarizes the relation between oxidative stress and T-cell function and the interaction between Th9 cells, interleukin-9 (IL-9) and oxidative stress in CLL. The T helper 9 cells have pleiotropic effects in anti-tumor immunity. They activate innate and adaptive anti-cancer immunity and induce tumor cell cycle arrest and apoptosis. However in CLL, IL-9 seems to participate in the pathogenesis of the disease by both enhancing proliferation and inhibiting apoptosis of cancer cells. Oxidative stress has been linked to the etiology and development of carcinogenesis. The redox state at the interface between Antigen Presenting Cell (APC) and T-cells within the immunological synapse also impacts on T-cell activation, proliferation and differentiation. Higher level of oxidative stress was observed in CLL patients that correlated with bad prognosis and the disease outcome. Oxidative stress can promote the activation of nuclear factor kappa-light-chain-enhancer of activated B cells (NFκB) which up regulates the expression of proinflammatory cytokines, including IL-9, which may be involved in the pathogenesis of CLL.
