In vitro Plant Regeneration from Cotyledon-derived Callus  of Three Varieties Pummelo (Citrus grandis [L.] Osb.)

Abstract: Cotyledon explants from in vitro grown seedlings were cultured on half-strength of MS medium with different growth regulators for in vitro indirect regeneration of shoots. Optimum callus formation, when cotyledon explants were cultured on MS containing 1.0 mg l-1 BAP with 5.0 mg l-1 NAA. After that the calli were used for shoot regeneration by transferring them  half-strength MS medium supplemented with only cytokinin. Maximum percentage of shoot regeneration was obtained on half strength MS medium in the presence of 1.0 mg l-1 BAP from callus in three varieties [Var.-1 (pink colour), Var.-2 (white colour) and Var.-3 (red colour)] of pummelo. For rooting, shoot cuttings were cultured on half strength MS salts with 0.1-1.0 mg l-1 NAA, IBA or IAA. The best and healthy rooting was observed on 0.1 mg l-1 NAA. The plantlets of three varieties were successfully established on soil. About 95% of plantlets survived under ex vitro condition.
