Evaluation of Some Factors Affecting Quantity, Quality and in vitro Maturation of Buffalo Oocytes

Abstract: This study aimed to evaluate the influence of collection techniques (dissection, aspiration, slicing and aspiration plus slicing) on yield and quality of buffalo oocytes in vitro matured (IVM) in TCM-199 or Ham’s F12 with or without hormones (follicle-stimulating hormone, FSH and estradiol, E2). Recovery Rate (RR) and category of the collected oocytes were determined and only Cumulus Oocyte Complexes (COCs) were matured in vitro. Results showed that RR was 84.6, 83.3, 72.7 and 52.0% for aspiration plus slicing, slicing, aspiration and dissection technique, respectively (p<0.05). Percentage of COCs was higher by slicing than aspiration, aspiration plus slicing and dissection (63.2 vs. 51.3, 51.2 and 42.0%, p<0.05). The corresponding percentages of expanded oocytes were 29.9, 30.3, 27.6 and 32.7%, respectively (p<0.05). Percentage of oocytes arrested at metaphase-II (MII) was higher (67.1%, p<0.001) while those at Germinal Vesicle (GV) (8.1%, p<0.001) and Deg. (9.1%, p<0.01) were lower in TCM-199 than in Ham’s F12. Hormonal addition increased percentage of oocytes arrested at metaphase-I (MI) and MII (p<0.05 and p<0.001, respectively) and decreased those at GV and Deg (p<0.001). Such results may indicate efficacy of slicing technique as a collection method on quantity and quality of buffalo oocytes. Also, in vitro maturation of buffalo oocytes was improved in TCM-199 supplemented with hormones (FSH and E2).
