Antimicrobial Activity of New Isolate Extract Against Bacterial Food Contaminant and Pathogen

Abstract:
The aims of this study were to screen and characterize Lactococcus lactis isolate that could produce a natural compound with a high antibacterial activity. Ninety one sample of raw cow milk from Sudan and 12 from Iraq were used in this study. The isolates were identified using morphological, physiological, biochemical characterizations, API system and molecular characterization. The inhibitory activity of the extract was evaluated against gram-negative bacteria (E. coli and pseudomonas aeruginosa) and gram positive bacteria (Bucillus subtilis, Staphylococcus aureus, Clostredium perfringens). The effect of pH, temperature and Ultra Violate (UV) on antimicrobial activity were optimized. Conventional, API system and molecular characterization confirmed that the new isolates were lactococcus lactis subsp. lactis. Their extract had a significant inhibitory effect on the pathogenic gram-negative and gram-positive strains. The optimum conditions of the extract were as follow: pH of 2.5, temperature of -4°C and at room temperature. The newly isolated Lactococcus lactis subsp. lactis produces a natural compound offering highly antimicrobial properties.
