Molecular Detection of NDM and VIM Genes in Pseudomonas aeruginosa Isolates from Clinical Samples in Bayelsa State, Nigeria

Abstract: Background and Objective: Pseudomonas aeruginosa is an organism widespread in nature and an important cause of healthcare-associated infection, especially in patients with compromised host defence. Acquired metallo-beta-lactamases (MBLs) pose a serious problem both in terms of treatment and infection control in the hospitals and reports across the world showed an increase in their prevalence, but the reports of the prevalence of MBLs are rare in Nigeria. This study aimed to detect the presence of NDM and VIM genes in Pseudomonas aeruginosa. Materials and Methods: A total of 146 clinical samples were collected. Samples collected were urine, sputum, endocervical swabs, high vaginal swabs and wound swabs. About 41.1% of the clinical samples were obtained from male patients while 58.9% were obtained from female patients. Phenotypic detection was carried out by the standard bacteriology method. The genomic DNA was extracted using the boiling method and amplification of genes was carried out using an AB19700 Applied Biosystem thermal cycler. Results: The total numbers of isolates obtained were 73. Bacteria isolates obtained were Escherichia coli, Klebsiella sp., Proteus spp., Staphylococcus aureus and Pseudomonas aeruginosa with 17 (23.3%), 15 (20.6%), 5 (6.8%), 20 (27.4%) and 16 (21.7%), respectively. Pseudomonas aeruginosa isolates obtained were highly susceptible to Streptomycin (93.8%) and high resistance to Nalidixic acid (56.25%). The VIM and NDM genes that were assayed for were not detected. The prevalence of NDM and VIM genes of Pseudomonas aeruginosa which poses a serious challenge for therapeutics and infection control is currently low in Bayelsa State, Nigeria. Conclusion: To restrict further spread, rational use of carbapenems and other antipseudomonal medications, regular surveillance and effective infection control measures should be implemented.
