Microencapsulation of Colistin Sodium Methanesulfonate in Gum Arabic and Maltodextrin by Spray Drying

Abstract:
The suitability of spray drying to encapsulate colistin sodium methanesulfonate in gum arabic in the presence of maltodextrin or sodium chloride was assessed with a view to generating microparticles that will not only retain anti-bacterial activity but might also be effective for inhalation therapy. The 300, 400 and 500 Units mL-1 colistin sodium methanesulfonate were separately spray dried in 2% gum arabic and 30% maltodextrin, 30% sodium chloride. The sensitivity of Escherichia coli NCIMB 8879 to the spray-dried particles was evaluated by the disc diffusion method to determine the residual activity of colistin sodium methanesulfonate. The size and shape of the spray-dried microparticles were determined by scanning electron microscopy and dynamic light scattering. Microparticles generated using gum arabic and sodium chloride did not inhibit the growth of the indicator organism whereas there was over 50% retention of activity of the 400 and 500 Units mL-1 colistin sodium methanesulfonate spray dried in gum arabic and maltodextrin compared with the controls. Scanning electron microscopy revealed that these later particles had smooth, non-uniform surfaces which confirm that encapsulation was successful. Furthermore, the particle size distribution under both conditions is also less than 10 μm. The results of this study suggest that spray drying can generate microparticles of colistin sodium methanesulfonate that retains antimicrobial activity and still satisfies the particle size requirement for effective inhalation therapy.
