Microscopic Studies on the Effect of Alprazolam (Xanax) on the Liver of Mice

Effects of the administration of Alprazolam (Xanax) were studied on the liver of the mice. Animals were treated orally with different daily doses of 0.01, 0.1 and 0.2 mg of alprazolam/kg body wt. for 21 days. In the animals treated with the two high therapeutic doses (0.1 and 0.2 mg), light microscopic examination showed enlargement of the nuclei, vacuolation of the cytoplasm, proliferation (hyperplasia) and hypertrophied of Kupffer cells. Extensive morphological changes in the liver cells were revealed by electron microscope including decrease in heterochromatin, extensive enlargement and fragmentation of the nucleoli, decrease of mitochondria, rough endoplasmic reticulum and glycogen contents, hypertrophied golgi apparatus, increase and dilation of smooth endoplasmic reticulum, increase of lipid droplets, vacuolation of cytoplasm.
