Cloning, Transformation and Localization of Thermotolerance ZmHSP22 Gene in Maize

Background and Objective: Plant heat shock proteins play an essential role in environmental stress tolerance. Maize small heat shock proteins were shown to accumulate to high levels during heat stress and play important role in the tolerance mechanism of plants to abiotic stresses. The purposes of the study were cloning, transformation and localization of the ZmHSP22 gene from an Egyptian line of maize (Zea mays L.). Materials and Methods: ZmHSP22 gene has been isolated, cloned and sequenced from the Egyptian maize line K1 after heat treatment at 45°C for 4 hrs. The gene was cloned in plant expression vector pCAMPIA1302 which produced a sub-cellular localization 22 kD protein. Transformation of the gene was performed after fusing with a green fluorescent protein and mitochondrial marker in leaves of the tobacco plant by the agroinfiltration method. Results: The isolated gene ZmHSP22 gave a size of 657 base pair encodes a protein of 218 amino acids. It has been submitted in the GenBank under accession number MT019963. The confocal fluorescence microscopy demonstrated that the gene ZmHSP22 expressed a high level of protein HSP22 accumulated in mitochondria during heat stress in the transgenic tobacco plants. Conclusion: ZmHSP22 gene encodes a mitochondrial protein HSP22 helps in protecting plant mitochondrial proteins against denaturation and degradation. ZmHSP22 gene will help to develop plant varieties adaptable to adverse environmental conditions.
