Molecular and Phylogenetic Study Based on Two Mitochondrial DNA Genes of Rabbit Pinworm “Passalurus ambiguus”

Background and Objectives: Passalurus ambiguus (Rudolphi, 1819) is one of the most prevalent intestinal nematodes of rabbits in Egypt and worldwide. The current work aimed to identify this common rabbit pinworm by molecular approach. Materials and Methods: The adult pinworms were collected from the large intestines of naturally infected rabbits. Passalurus ambiguus-genomic DNA was extracted. Amplification of partial cytochrome oxidase 1 (cox1) and cytochrome b (cyt b) genes of mitochondrial DNA (mt DNA) was carried out using the following primer sets: for cox1 5-TTATGCCTATTATGATG-3 (Forward), 5-AACAACCAACTAAAAAC-3 (Reverse) and for cyt b gene 5-TATTATGGGACTGATTT-3 (Forward), 5-ATAACAACACCTTTACA-3 (Reverse). DNA extraction, amplification and sequencing of the 2 selected genes were done successfully. Results: Comparing of the 2 mtDNA genes showed higher similarity (98-99%) with those deposited in databases for P. ambiguus isolates from other parts of the world, thus confirming the identity of the species in Egypt. Conclusion: This study is in the way of revealing the genetic makeup of pinworms of family Oxyuridae that might help in better identification of this group of helminths that have both veterinary and public health importance. The current study proved that mtDNA genes can be a suitable target for identification and discrimination of different pinworms.
