| Comparative Effect of Inhibitors on the ATPases from the Excretory System of the Usherhopper, Poekilocerus bufonius and Desert Locust, Schistocerca gregaria |

Abstract:
The bafilomycin A1 and N-ethylmaleimide (NEM)-sensitive (V-type) ATPase was partially purified from the apical membranerich fractions of excretory system (Malpighian tubules and hind gut) of Poekilocerus bufonius and Schistocerca gregaria. Enzymatic activity was inhibited by bafilomycin A1 (IC50 = 0.48 and 1.3 μM for S. gregaria and P. bufonius, respectively) and NEM (IC50 = 10.1 and 2.8 μM of P. bufonius and S. gregaria, respectively). The V-type ATPase activity is confined to the apical membrane fraction, while the activity of Na+/K+ -ATPase forms the major part of the basal membrane fraction of both insects. The chloride salts also caused an increase in activity in the following ascending order: RbCl, LiCI, choline Cl, NaCI, KCl and tris-HCl of both insects. The present results show that the properties of V-type ATPase of P. bufonius and S. gregaria are similar to those reported for other insect tissues. |
